1. Detection and Structure Elucidation of Drug Metabolites in Biological Samples using HPLC-MS/MS Techniques
Sammanfattning : This thesis describes the structure elucidation of drug metabolites in biological samples by the use of high performance liquid chromatography (HPLC) atmospheric pressure ionization (API) tandem mass spectrometry (MS/MS). Due to their different advantages, various mass analyzers have been used in the different experiments. LÄS MER
2. Porphyrins and heme in microorganisms : Porphyrin content and its relation to phototherapy and antimicrobial treatments in vivo and in vitro
Sammanfattning : One of the greatest threats to human health is increasing antimicrobial resistance among pathogens, and finding alternatives for treatment of bacterial infections is of highest importance together with a more controlled use of antibiotics. Porphyrins and heme have both been shown to be a promising class of compounds for inactivation of bacteria; porphyrins by their excellent properties to act as a photosensitizer, and heme by its importance as an iron source during a bacterial infection in vertebrates. 3. Mass Spectrometric Analysis of Oxylipins : Application to Cytochrome P450-Dependent Metabolism
Sammanfattning : Cytochrome P450 (CYP) family 4 constitutes monoxygenases responsible for hydroxylation of fatty acids and other lipids. For example, CYP4F3 metabolizes leukotrienes and CYP4F8 prostaglandin H. Importantly, six of the twelve CYP4 enzymes are orphans, i.e. LÄS MER
4. The structural basis for the catalytic specificity of manganese lipoxygenases : 3D structure analysis of the lipoxygenase of Magnaporthe oryzae
Sammanfattning : Lipoxygenases (LOX) catalyze regio- and stereospecific oxygenation of polyunsaturated fatty acids to hydroperoxides. These hydroperoxides are further metabolized to leukotrienes and lipoxins in mammals, and are involved in asthma and inflammation. LOX of animals and plants contain iron as catalytic metal (FeLOX). 5. The in-vitro and in-vivo metabolism of the oral anticoagulant phenprocoumon as influenced by genetic polymorphisms of cytochrome P4502C9
Sammanfattning : Oral anticoagulants are widely used for the prevention of thromboembolic disorders. Warfarin (WA) is most commonly used world-wide, while phenprocoumon (PPC) is the first-line anticoagulant in some European countries including Germany. Each anticoagulant exists in two different enantiomeric forms and is administered orally as a racemate. LÄS MER
