1. Down syndrome : Growth and endocrine impact
Sammanfattning : Down syndrome (DS) is associated with psychomotor retardation, short stature and endocrine dysfunction. Statural growth is a well-known indicator of health. The growth in DS differs markedly from that of other children and there is a 20 cm reduction of final height as compared to target height. LÄS MER
2. Cardiovascular effects of growth hormone, IGF-I and growth hormone secretagogues
Sammanfattning : Growth hormone (GH) may exert direct growth-promoting and metabolic actions on target tissues, but most of its effects are mediated by circulating (endocrine) or local (auto-/paracrine) insulin-like growth factor-I (IGF-I). The GH/IGF-I system has an important role during cardiac development and for maintaining the structure and function of the heart. LÄS MER
3. Growth hormone responsiveness in children : results from Swedish multicenter clinical trials of growth hormone treatment
Sammanfattning : The general aims of the thesis were to study GH responsiveness by estimation of pharmacokinetics and bioavailability of injected recombinant human GH (rhGH), of growth response as gain in heightSDS during childhood and puberty, and IGF-I response as change in circulating IGF-ISDS and IGFBP3SDS. Methods Short children were recruited during 1988–1999 into two national randomized multicentre clinical trials on growth until adult height. LÄS MER
4. Metabolic responsiveness to growth hormone in children
Sammanfattning : Metabolic effects of growth hormone (GH) therapy in short children have not been clearly established owing to the previous lack of controlled trials studying the metabolic outcome in response to different GH doses despite the known effects of GH on insulin sensitivity, lipid profile, and body composition. It has previously been shown that individualized GH doses during catch-up growth significantly reduce the proportion of unexpectedly good and poor responders around a predefined individual growth target in short prepubertal children. 5. Identification of novel growth hormone-regulated factors
Sammanfattning : The studies described in this thesis aimed to identify novel factors involved in the regulation of longitudinal growth and bone mineralization in response to growth hormone (GH) treatment. This was done by performing a single factor study (Paper I) where it was found that growth response was negatively correlated with adiponectin levels during the first year of GH treatment in short prepubertal children.
