  1. 1. Protein Folding, Binding and Evolution : PDZ domains and paralemmins as model systems

    Författare :Greta Hultqvist; Per Jemth; Ruth Nussinov; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Protein folding; evolution; binding; allostery; Paralemmin; PDZ domain;

    Sammanfattning : Proteins present at the synapse need to be multitasking in order to perform all vital functions in this limited space. In this thesis I have analyzed the function and evolution of such proteins, focusing on the PDZ domain and the paralemmin family. The PDZ domains bind to a wide variety of interaction partners. 2. 2. Design, expression, and analysis of antibody-based blood-brain barrier shuttles

    Författare :Andrés de la Rosa; Greta Hultqvist; Paul O'Callaghan; Nicole G. Metzendorf; Torben Moos; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Antibody therapeutics; transient gene expression; protein production; blood-brain barri-er BBB ; BBB shuttle; brain delivery; Transferrin receptor TfR ; heparan sulfate; holo-transferrin; Molekylär biovetenskap; Molecular Life Sciences;

    Sammanfattning : Antibody therapeutics, with their strong and highly selective target binding, are now used to treat various diseases. However, to enable their use to treat brain disorders, they must be delivered across the blood-brain barrier (BBB), as without active transport, only around 0.01% of intravenously injected doses reach the brain. 3. 3. Preclinical PET imaging and therapy of Alzheimer's disease

    Författare :Silvio R. Meier; Stina Syvänen; Dag Sehlin; Greta Hultqvist; Matthias Brendel; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer s disease; amyloid-beta; positron emission tomography PET ; bispecific antibody; BACE-1; TREM2; GFAP; Molekylär medicin; Molecular Medicine; Neuroscience; Neurovetenskap;

    Sammanfattning : The main histopathological hallmarks of Alzheimer’s disease are extracellular amyloid-beta (Aβ) plaques and intracellular neurofibrillary tangles, containing tau protein. Because of misfolded and aggregated proteins, activated microglia and astroglia react with a neuroinflammatory response, which may contribute to disease progression and severity. LÄS MER

  4. 4. Enhancing the therapeutic effect of biological drugs with protein engineering : Focusing on pre-clinical Alzheimer’s disease therapy

    Författare :Fadi Rofo; Greta Hultqvist; Stina Syvänen; Sara Mangsbo; Joy Zuchero; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer’s disease; amyloid-beta; blood-brain barrier; transferrin receptor; protein-drugs; multivalent-bispecific antibodies;

    Sammanfattning : Aggregation of the amyloid-β peptide (Aβ) is one of the main pathological hallmarks in Alzheimer’s disease (AD). The soluble Aβ aggregates (oligomers and protofibrils) have shown to be the most harmful species. Hence, targeting these aggregates can be of therapeutic potential. LÄS MER
