22. Restorative and neuroprotective treatment strategies for Parkinson’s disease: tudies on fetal dopamine cell transplants and GDNF delivery by intracerebral injections and recombinant AAV vectors
Sammanfattning : The intrastriatal 6-hydroxydopamine lesion model in rats is particularly useful in studies testing new therapeutic strategies for Parkinson’s disease. This thesis work (1) describes the characterization of the motor deficits after different types of intrastriatal lesions and identifies the four-site terminal lesion as a model of symptomatic Parkinson’s disease; (2) using the same model, provides evidence that the functional impact of the dopamine cell grafts are dependent on interactions with the host brain; (3) demonstrates that protection or restitution of normal motor function after 6-hydroxydopamine lesions can be achieved if glial cell line-derived neurotrophic factor is administered in the striatum at the time of the terminal fiber degeneration. 24. Intracranial and intraocular grafting of fetal ventral mesencephalic and striatal tissues : neuronal survival and nerve growth characteristics
Sammanfattning : Parkinson's disease is caused by degeneration of dopaminergic neurons in the ventral mesencephalon. Various animal models and clinical trials has shown that grafts of fetal ventral mesencephalic tissue can survive and functionally reinnervate the dopamine-depleted striatum, leading to long-term improvements of therapeutic value, but still symptomatic relief is far from complete. 25. Spinal cord grafts : interaction with cografts and effects of trophic factors
Sammanfattning : Spinal cord injury is common in the modern society, often afflicts young individuals, and often has devastating consequences. The present work studies possibilities to obtain better conditions for regeneration of the transected spinal cord in rats, conditions under which spinal cord grafts are able to establish structural and functional connections with "natural" targets, and effects of a series of neurotrophic factors on spinal cord graft survival and growth.
