  1. 1. Intracellular unbound drug concentrations : Methodology and application for understanding cellular drug exposure

    Författare :André Mateus; Per Artursson; Pär Matsson; José A. G. Morais; Jeff Krise; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intracellular unbound drug concentrations; free drug; drug binding; drug transport; drug accumulation; cellular drug response; drug target engagement; Pharmaceutical Science; Farmaceutisk vetenskap;

    Sammanfattning : Most known drug targets and metabolizing enzymes are located inside cells. Interactions with these proteins are determined by intracellular unbound drug concentrations. Assessing intracellular drug exposure is technically challenging, but essential for predicting pharmacokinetic, pharmacological, and toxicological profiles of new drugs. LÄS MER

  2. 2. Characterization of parameters influencing intracellular bioavailability and prediction of intracellular drug exposure

    Författare :Andrea Treyer; Per Artursson; Gert Fricker; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; intracellular drug bioavailability; unbound drug concentration; drug disposition; ADME; drug transport; drug metabolism membrane partitioning; phospholipid; drug-drug interaction; antisense oligonucleotide; cell-penetrating peptide; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : This thesis work investigates factors influencing intracellular drug disposition. An experimental method for measurement of intracellular bioavailability (Fic), was used throughout. Fic is defined as the ratio between the unbound drug concentration inside the cell and the compound concentration in the cell exterior. LÄS MER

  3. 3. Towards specific quantification of exosomes using surface-based sensing

    Författare :Deborah Rupert; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; label-free; surface plasmon resonance.; exosome; binding rate; antibody; concentration; nanoparticle tracking analysis; quantification; liposome; cluster of differentiation 63 CD63 ; diffusion; specific detection; mass transport limited conditions; extracellular vesicles; surface-based biosensor;

    Sammanfattning : When exosomes were first observed in 1967, they were believed to be cell debris until extensive studies demonstrated key biological functionalities related to their composition and formation mechanism. Exosomes are nanometer-sized lipid vesicles of endocytic origin, with a membrane consisting of lipids and proteins and encapsulating a cargo composed of soluble proteins and genetic material. 4. 4. Miniaturized, Membrane-Based and Environmentally Green Sample Preparation Systems: Applications for Biological and Environmental Aqueous Samples

    Författare :Thaer Barri; Centrum för analys och syntes; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Analytisk kemi; Analytical chemistry; Free concentration; Exhustive extraction; Microporous membrane liquid-liqid extraction; Miniaturization; Supported liquid membrane extraction; Environmental chemistry; Miljökemi;

    Sammanfattning : Contemporary advancements in miniaturization of analytical systems have advantageously fostered development, automation, and hyphenation of a variety of sample preparation techniques. Among several downscaled extraction designs, porous synthetic polymeric membranes (such as flat sheet (FS) or hollow fiber (HF)) have been utilized for membrane-based extraction, separating two aqueous phases (as in supported liquid membrane (SLM) extraction or one aqueous phase and another organic phase (as in microporous membrane liquid-liquid extraction (MMLLE)). 5. 5. Mass spectrometry based analysis of drugs, neurotransmitters and lipids in invertebrate model systems

    Författare :Nhu TN Phan; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; CE-MS; Imaging mass spectrometry; Lipids; Neurotransmitters; Drug; Drosophila; C. elegans;

    Sammanfattning : Mass spectrometry (MS) is one of the most universal analytical techniques due to its label-free detection principle and high chemical specificity, high selectivity, and sensitivity. MS is diverse with many different types of systems to meet different analytical demands from various research areas. LÄS MER
