  1. 6. Distribution and Chemical Diversity of Cyclotides from Violaceae : Impact of Structure on Cytotoxic Activity and Membrane Interactions

    Författare :Robert Burman; Ulf Göransson; Lars Bohlin; William Gerwick; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cyclotide; Violaceae; Viola; Cytotoxic; Mechanism; In vivo; Structure-activity; Membrane; Mass spectrometry; Pharmacognosy; Farmakognosi; Pharmacognosy; Farmakognosi;

    Sammanfattning : During the last decade there has been increased interest in the cyclotide protein family, which consist of a circular chain of approximately 30 amino acids, including six cysteines that form three disulfide bonds, arranged in a cyclic cystine knot motif. This thesis gives new insights in cyclotide distribution and occurrence in the plant family Violaceae, structure-activity relationships for cytotoxic effects, membrane disruption and adsorption on lipid membranes, and evaluates toxicity and anti-tumor activity in vivo. 2. 7. Studies of Cytotoxic Compounds of Natural Origin and their Mechanisms of Action

    Författare :Jenny Felth; Lars Bohlin; Joachim Gullbo; Ulf Göransson; Thomas Efferth; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; cytotoxic; cardiac glycoside; gambogic acid; cancer; mechanism of action; Pharmacognosy; Farmakognosi; Pharmacognosy; Farmakognosi;

    Sammanfattning : Cancer incidence is increasing and novel anticancer drugs with new mechanisms of action are essential for future chemotherapeutic treatment. Natural products have historically played an important role in the development of anti-cancer drugs and have potential to do so also in the future. LÄS MER

  3. 8. Bioactive Compounds in the Chemical Defence of Marine Sponges : Structure-Activity Relationships and Pharmacological Targets

    Författare :Erik Hedner; Lars Bohlin; Per R Jonsson; Raffaele Riccio; Uppsala universitet; []
    Nyckelord :Pharmacognosy; 5-hydroxytryptamine; agelasine; agelasimine; antibacterial; antifouling; barettin; bromobenzisoxazolone barettin; cytotoxic; marine; secondary metabolite; sponge; Farmakognosi; Agelas; Geodia barretti;

    Sammanfattning : Marine invertebrates, in particular sponges, represent a source of a wide range of secondary metabolites, many of which have been attributed various defensive capabilities against environmental stress factors. In this thesis sponge-derived low-molecular peptide-like compounds and associated analogs are investigated for bioactivity and pharmacological targets. LÄS MER

    Sammanfattning : This thesis investigates the anti-leukemic activity of terpenoids isolated from medicinal mushrooms and marine sponges, as well as their possible targets and mechanisms of action.In the first section, we focused on studying the triterpenoidal components of three triterpenoid-enriched medicinal mushrooms Antrodia cinnamomea, Ganoderma lucidum, and Poria cocos, which have been used in folk medicine for centuries and also developed into several contemporary marketed products.
