1. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury
Sammanfattning : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 2. Hepatic 3D spheroids as a model system for drug-induced liver injury and cytochrome P450 induction
Sammanfattning : Drug-induced liver injury (DILI) is a major cause of post-marketing drug withdrawals and restricted-use warnings. In addition, unexpected adverse hepatic drug reactions occurring in the clinical phases of development are a major reason for drug attrition. LÄS MER
3. Array-based identification of disease-associated proteins
Sammanfattning : To increase our understanding of the human body in both health and disease, proteins can be studied in samples such as plasma and serum to provide a molecular profile of the physiological status. In the work presented in this thesis, array-based methods were used to study associations of protein and autoantibody profiles with disease. LÄS MER
4. Endotoxin-, glutamate- and drug-induced inflammation and cytotoxicity with emphasis on signal transduction mechanisms
Sammanfattning : Inflammatory cells such as monocytes and glial cells play an important role in the pathogenesis of ischemic- or drug-induced brain or liver injury. Excessive concentrations of glutamate is toxic to neural cells, and triggers a series of transcriptional events beginning with the expression of the immediate early genes like c-fos and c-jun, which in turn affect the expression of other genes necessary for the development of brain injury. 5. Non-Invasive Characterization of Liver Disease : By Multimodal Quantitative Magnetic Resonance
Sammanfattning : There is a large and unmet need for diagnostic tool that can be used to characterize chronic liver diseases (CLD). In the earlier stages of CLD, much of the diagnostics involves performing biopsies, which are evaluated by a histopathologist for the presence of e.g. fat, iron, inflammation, and fibrosis. LÄS MER
