  1. 1. Fragment-based drug discovery : Novel methods and strategies for identifying and evolving fragment leads

    Författare :Edward A. FitzGerald; Helena Danielson; Hanna-Kirsti Schrøder Leiros; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Drug Discovery; Biophysics; Fragment-based drug discovery; Epigenetics; Biosensors; Surface Plasmon Resonance; Interaction Analysis; Second-Harmonics; Biokemi; Biochemistry;

    Sammanfattning : The need for new drugs became ever more apparent in the year 2020 when the world was faced with a viral pandemic. How drugs are discovered and their relevance to society became part of daily discussions in workplaces and homes throughout the world. Consequently, efficient strategies for preclinical drug discovery are clearly needed. LÄS MER

    Sammanfattning : In 2001 the human proteome organization (HUPO) was created with the ambition to identify and characterize all proteins encoded in the human genome according to several criteria; their expression levels in different tissues and under different conditions; the sub-cellular localization; post-translational modifications; interactions, and if possible also the relationship between their structure and function.When the knowledge of different proteins and their potential interactions increases, so does the need for methods able to unravel the nature of molecular processes in cells and organized tissues, and ultimately for clinical use in samples obtained from patients. 3. 3. Interaction Studies of Secreted Aspartic Proteases (Saps) from Candida albicans : Application for Drug Discovery

    Författare :Dan Backman; Helena Danielson; Birgitta Tomkinson; Gunnar Henriksson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; SPR Biosensor; Secreted aspartic proteases; Candida albicans; interaction kinetics; drug discovery; protease inhibitor; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : This thesis is focused on enzymatic studies of the secreted aspartic proteases (Saps) from Candida albicans as a tool for discovery of anti-candida drugs. C. albicans causes infections in a number of different locations, which differ widely in the protein substrates available and pH. Since C. LÄS MER

  4. 4. Towards a New Generation of Anti-HIV Drugs : Interaction Kinetic Analysis of Enzyme Inhibitors Using SPR-biosensors

    Författare :Malin Elinder; U. Helena Danielson; Maurizio Botta; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; drug discovery; fragment; screening; reverse transcriptase; microbicides; interaction analysis; NNRTIs; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Sammanfattning : As of today, there are 25 drugs approved for the treatment of HIV and AIDS. Nevertheless, HIV continues to infect and kill millions of people every year. Despite intensive research efforts, both a vaccine and a cure remain elusive and the long term efficacy of existing drugs is limited by the development of resistant HIV strains. LÄS MER

  5. 5. Structure and Function of Aqua(glycero)porins - A Path to Drug Design

    Författare :Gerhard Fischer; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; structural biology; drug screening; Malaria; membrane proteins; yeast;

    Sammanfattning : Membrane proteins are a major class of the drug targets. Rational design of drugs requires knowledge about the structure and function of these proteins. Working with membrane proteins, however, is experimentally challenging as the required technology is still emerging. LÄS MER
