  1. 11. Ancient genomics of Baltic seals : Insights on the past Baltic grey seal and harp seal populations

    Författare :Maiken Hemme Bro-Jørgensen; Kerstin Lidén; Morten Tange Olsen; Aikaterini Glykou; Olle Karlsson; Stockholms universitet; []
    Nyckelord :HUMANIORA; HUMANITIES; Ancient DNA; Baltic Sea; DNA preservation; extinction; mitogenome; seal hunting; sex identification; arkeologi med inriktning mot laborativ arkeologi; Scientific Archaeology;

    Sammanfattning : This thesis aims to study and describe the ancient populations of grey and harp seals in the Baltic Sea, and to present new methodological approaches for general use in ancient DNA studies.The dissertation is comprised of five studies: a review of the use of paleogenetics in studying ancient human-marine mammal interactions; a method paper investigating patterns of DNA preservation in ancient pinniped samples; a method paper presenting a genetic sex identification method for ancient pinnipeds; a population genomic study of the Baltic grey seal; and a population genomic study of the now extinct Baltic harp seal. 2. 12. Single Molecule Detection : Microfluidic Automation and Digital Quantification

    Författare :Malte Kühnemund; Mats Nilsson; Ulf Landegren; Sten Linnarsson; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; single molecule; digital; rolling circle amplification; magnetic particle; padlock probe; microfluidics; resistive pulse sensing; lab on chip; mobile phone microscopy; enrichment; sequencing;

    Sammanfattning : Much of recent progress in medical research and diagnostics has been enabled through the advances in molecular analysis technologies, which now permit the detection and analysis of single molecules with high sensitivity and specificity. Assay sensitivity is fundamentally limited by the efficiency of the detection method used for read-out. LÄS MER

  3. 13. Quantitative detection of bacterial DNA in whole blood in bloodstream infection

    Författare :Ingrid Ziegler; Kristoffer Strålin; Paula Mölling; Magnus Rasmussen; Örebro universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Bloodstream infection; bacteremia; sepsis; DNA load; quantitative PCR; droplet digital PCR; 16S rDNA;

    Sammanfattning : This thesis aims to increase the knowledge on how quantitative PCR can be used in the diagnostics of bloodstream infections, with an emphasis on quantitative elements.In Papers I and II, we evaluated quantitative data from two commercial PCR tests for pathogen detection directly in blood, Magicplex Sepsis (I) and SeptiFast (II), from patients with suspected sepsis. 4. 14. Ultrasensitive DNA sequencing using liquid biopsies enables precision medicine

    Författare :Gustav Johansson; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Liquid biopsy; cell-free DNA; immune repertoire; next-generation sequencing; unique molecule identifier; GIST; γδ T cell;

    Sammanfattning : Liquid biopsies are minimally invasive and allow repetitive sampling of body fluids. Analysis of cell-free tumor DNA in liquid biopsies can be used as a biomarker for cancer. However, in most clinically relevant liquid biopsies, cell-free DNA is present at low concentrations, contains minute tumor allele frequencies, and is highly fragmented. LÄS MER

  5. 15. Hepatitis B virus RNA in serum and liver tissue - quantification using digital PCR

    Författare :Kasthuri Prakash; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; hepatitis B virus; NTCP; HBV DNA; HBV RNA; subviral particles; integrations; droplet digital PCR;

    Sammanfattning : Hepatitis B virus (HBV) infection is a global health issue that is responsible for approximately 900,000 deaths each year, by inducing liver cirrhosis and hepatocellular carcinoma (HCC). A few markers are used to classify HBV infection and monitor treatment efficacy, including HBV DNA, surface antigen (HBsAg) and e antigen (HBeAg) in serum as well as HBV DNA and RNA in liver tissue.
