    Sammanfattning : It is becoming increasingly clear that long-range control of gene expression is mediated through direct physical interactions between genes and regulatory elements, either intra- or interchromosomally. In addition to transcriptional initiation, formation of active chromatin hubs seem to be crucial for increased transcriptional efficiency as well as insulation from neighbouring heterochromatic environment.     Sammanfattning : The theme of this thesis has been chromatin organization ranging from methodological studies to involvement in apoptotic response. The aims have been: (i) to compare the information obtained by the AVTD method and comet assay concerning DNA-loop organization, (ii) to test the hypothesis that DNA-loop relaxation could be the triggering signal for induction of apoptosis in G0-lymphocytes, and (iii) to study the dose response for inhibition of the replication fork movement and pathways for the DNA repair process at the replication fork.     Sammanfattning : A human genome laid out in a straight line would measure 2m from end to end, but in living cells it is folded into a compact structure contained in a nuclear space with a diameter of 2µm. This compact genome is hierarchically organized and spatiotemporally regulates distinct cellular gene expression mechanisms via promoter - enhancer chromatin loops. LÄS MER

    Sammanfattning : Ribosomal biogenesis and protein translation are finely coordinated with cell proliferation. All three RNA polymerases Pol I, II, and III are utilized for highly efficient and accurate ribosome production. The transcriptional activity of Pol I has been found to be a key determinant for ribosome biogenesis. LÄS MER
