  1. 1. Modeling Human Hematopoiesis Using the CRISPR/Cas9 System

    Författare :Alexandra Bäckström; Avdelningen för molekylärmedicin och genterapi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Hematopoiesis; Hematopoietic stem and progenitor cells; HSC regulation; CRISPR Cas9; Gene editing;

    Sammanfattning : Hematopoietic stem cells (HSCs) have the ability to self-renew and to give rise to all blood cells of the different lineages, and are thereby responsible for the replenishment of blood cells throughout life. These cells are tightly regulated by extrinsic and intrinsic regulators, such as signals from the bone marrow microenvironment, complex transcription factor networks and epigenetic regulators. 2. 2. Multifaceted RNA-mediated regulatory mechanisms in Streptococcus pyogenes

    Författare :Anaïs Le Rhun; Emmanuelle Charpentier; Franz Narberhaus; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Streptococcus pyogenes; small RNAs; CRISPR; Cas9; tracrRNA; RNases; gene expression; RNA sequencing; Infectious Diseases; infektionssjukdomar; mikrobiologi; Microbiology;

    Sammanfattning : Bacterial pathogens rely on precise regulation of gene expression to coordinate host infection processes and resist invasion by mobile genetic elements. An interconnected network of protein and RNA regulators dynamically controls the expression of virulence factors using a variety of mechanisms. LÄS MER

  3. 3. Improving integration efficiency and precision of CRISPR/Cas9-mediated genome editing

    Författare :Sandra Wimberger; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Genome editing; CRISPR-Cas9; DNA repair; genomic integration; off-targets;

    Sammanfattning : The clustered regularly interspaced short palindromic repeats (CRISPR) – CRISPR-associated protein 9 (Cas9) system has revolutionized the field of genome engineering, providing a cost-effective and fast tool for targeted gene modifications. Endogenous repair pathways, including error-prone non-homologous end joining and alternative end joining, or precise homology-directed repair (HDR), mend Cas9-induced DNA double-strand breaks. 4. 4. CRISPR/Cas9-based therapies and the role of astrocytes in Alzheimer’s disease and Parkinson’s disease

    Författare :Evangelos Konstantinidis; Martin Ingelsson; Anna Erlandsson; Vilmantas Giedraitis; Magnus Essand; Harald Steiner; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Alzheimer s disease; Parkinson s disease; gene editing; CRISPR Cas9; amyloid-beta; astrocytes; neurons; iPSCs; electrophysiology;

    Sammanfattning : Alzheimer’s disease (AD) and Parkinson’s disease (PD) are the two most common neurodegenerative disorders. Whereas the AD brain features plaques of amyloid-beta (Aβ) and neurofibrillary tangles of tau, the PD brain is characterized by Lewy bodies and Lewy neurites containing α-synuclein (αSyn). LÄS MER

  5. 5. Exploring early development and regenerative medicine using CRISPR/Cas9

    Författare :Nerges Winblad; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : With their intrinsic capacity to self-renew and their potential to differentiate to specialized tissues, human embryonic stem cells (hESCs) have many applications in the fields of cell therapy and developmental biology. By applying clustered regularly interspaced short palindromic repeats/CRISPR-associated protein 9 (CRISPR/Cas9) genome engineering on selected genes in hESCs, we are able to create cells with desired characteristics.
