11. Studies on DNA adducts and carcinogenesis with 2-nitrofluorene as a model substance
Sammanfattning : A number of carcinogens or their electrophilic intermediates are able to bind covalently to DNA, forming DNA adducts. DNA adducts may cause gene mutations if they are misrepaired or remain unrepaired before DNA replication occurs. DNA adduct formation is believed to be the initial step of chemical carcinogenesis. 12. Mortality and morbidity in lead smelter workers with concomitant exposure to arsenic
Sammanfattning : Arsenic is a well-known lung carcinogen in humans. In 2006, IARC upgraded inorganic lead as a possible human carcinogen (2A). LÄS MER
13. Improved basis for cancer risk assessment of acrylamide from food : Determination of glycidamide in vivo doses
Sammanfattning : Acrylamide is formed in heat processing of many common foods. According to animal cancer tests acrylamide is a carcinogen. To estimate the cancer risk from exposure via food, the response at high doses in the cancer tests with rats has to be extrapolated to the exposure levels in humans. LÄS MER
14. The attenuation of the P53 response to DNA damage in rodent liver preneoplastic enzyme-altered foci
Sammanfattning : In connection with life-time bioassays in rodents, the liver is one of the organs most frequently affected. During the carcinogenic process, single preneoplastic hepatocytes develop into hepatocellular adenoma or carcinoma (HCC). LÄS MER
15. Adrenal Bioactivation and Toxicity of 3-MeSO2-DDE, o,p´-DDD and DMBA Investigated in Tissue Slice Culture
Sammanfattning : I developed a precision-cut adrenal slice culture procedure to investigate cytochrome P450 (CYP) catalysed irreversible binding and adrenocorticolytic effects in human, rodent, and fish adrenal tissue, ex vivo. Autoradiography and radioluminography of exposed tissue slices showed that the potent adrenal toxicant 3-methylsulphonyl-2,2´-bis(4-chlorophenyl)-1,1´-dichloroethene (MeSO2-DDE) causes a selective metabolite binding in zona fasciculata (ZF), which is diminished by the CYP11B1 inhibitor metyrapone.
