  1. 16. Variation and comparison of cytochromes P450 in human liver and blood : with special emphasis on gene expression

    Författare :Niklas Finnström; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Cytochrome P450; CY1A2; CYP1B1; CYP2E1; CYP3A4; gene expression; RTPCR; liver; blood; human; variation.;

    Sammanfattning : The liver is the major drug and xenobiotic metabolising organ, and is the target of many adverse reactions. The main hepatic metabolic enzymes belong to the mono oxygenase enzyme family of cytochromes P450 (CYP). Direct studies of these enzymes in small liver biopsy samples require ultrasensitive methodology. LÄS MER

  2. 17. In vitro and in vivo studies of artemisinin endoperoxides

    Författare :Therese Ericsson; Göteborgs universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Artemisinin; Artesunate; Breast cancer; Cytochrome P450; Dihydroartemisinin; Inhibition; LC-MS MS; Pharmacokinetics; Plasma; Saliva;

    Sammanfattning : Artemisinin and its semi-synthetic derivatives (eg. Artemether/ARM, artesunate/ARS, dihydroartemisinin/DHA) play an important role in combating malaria, and treatments containing an artemisinin derivative (artemisinin-based combination therapies, ACTs) are today the standard treatment worldwide for Plasmodium falciparum malaria. 3. 18. Extrahepatic cytochrome P450s : relation to cancer susceptibility

    Författare :Tove Rylander Rudqvist; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Cytochrome P450 (CYP) enzymes in extrahepatic tissues may play important roles in regulating the capacity of individual cells to metabolize environmental carcinogens and hormones. In this thesis, we characterized the cytochrome P450 profile of the human normal breast and investigated whether polymorphisms in the genes encoding the estrogen metabolizing enzymes CYP1131 and Catechol-O-methyltransferase (COMT) were associated with breast or endometrial cancer risk. 4. 19. Pharmacogenetic and pharmacokinetic studies of cyclophosphamide : in cell, animal and human

    Författare :Hanjing Xie; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Cyclophosphamide; Cytochrome P450; Metabolism; Pharmacokinetics; 4Hydroxycyclophosphamide;

    Sammanfattning : Cyclophosphamide (CPA) is widely used anti-cancer drug that has to be catalyzed by the cytochrome P450 (CYP) enzyme 2B6 as well as some other CYPs to exert its cytotoxic activity. A high degree of interindividual variation in CPA kinetics has been reported in both children and adult patients, which may be due to several factors such as concomitantly used drugs, variation in physiological status of individual patients and genetic polymorphism of the CYP enzymes. 5. 20. Electrochemical and Enzymatic In Vitro Studies on Reactive Drug Metabolites. Synthesis, Characterization and Avoidance

    Författare :Tove Johansson Mali´n; Göteborgs universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Cytochrome P450; Drug; Electrochemical oxidation; Electrochemistry; Intermediate; Metabolism; Metabolic activation; Mimicry; Reactive Metabolite; Trapping.;

    Sammanfattning : During development of pharmaceuticals, it is essential to as early as possible identify and preferably avoid formation of reactive metabolites and intermediates. Reactive metabolites may represent obstacles in the development and use of drugs, due to their potential toxicity. LÄS MER
