  1. 1. The phencyclidine model of schizophrenia : dysregulation of brain dopamine systems induced by NMDA receptor antagonists

    Författare :Jan M Mathé; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :phencyclidine; MK-801; CNQX; prazosin; tetrodotoxin; dopamine; neurotensin; ventral tegmental area; nucleus accumbens; medial prefrontal cortex;

    Sammanfattning : Non-competitive NMDA receptor antagonists, such as phencyclidine (PCP) and dizocilpine (MK-801) can even acutely induce a drug induced psychotic state which closely resembles schizophrenia. The midbrain dopamine (DA) neurons, which originate in the ventral tegmental area (VTA), are involved in motivational, attentional and cognitive processes, and have also been profoundly implicated in schizophrenia. 2. 2. Microfluidics based techniques for electrophysiological studies of cells

    Författare :Aikeremu Ahemaiti; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES;

    Sammanfattning : This thesis work investigates the application of microfluidics to perform electrophysiological studies on cells, including investigations of the effect of cholesterol on the dynamic ion permeability of TRPV1 ion channels, and the application of a microfluidic device, the multifunctional pipette, in electrophysiological studies on brain slices. In the first part of this thesis, Chinese hamster ovary (CHO) cells overexpressing the TRPV1 ion channel were used in a dynamic ion permeability study, where the activation properties of the TRPV1 ion channel were investigated using the patch clamp technique after depletion of membrane cholesterol. 3. 3. Molecular studies of the insulin-like growth factor system in the brain : focus on growth, tumorigenesis and trauma

    Författare :Ann-Christin Sandberg Nordqvist; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :insulin-like growth factors; insulin-like growth factor binding proteins; brain; mRNA; meningioma; glioma; traumatic brain injury; in situ hybridization; Northern blothybridization;

    Sammanfattning : Insulin-like growth factor (IGF) 1 and -2 are synthesized by most tissues of the body and function as autocrine or paracrine agents in a tissue-specific manner. mRNA for IGF-1 and -2, as well as the IGF-binding proteins, are synthesized locally in various regions of the brain. 4. 4. Afferent input to midbrain dopamine neurones and its modulation : an electrophysiological study i vitro

    Författare :Pernilla Grillner; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :dopamine neurones; synaptic transmission; NMDA receptors; L-type Ca2 channels; presynaptic inhibition; metabotropic glutamate receptors; muscarine; nicotine;

    Sammanfattning : Dopamine (DA) neurones of the ventral mesencephalon are involved in the control of reward related behaviour, cognitive functions and motor performances, and provide a critical site of action for major categories of neuropsychiatric drugs, such as antipsychotic agents, dependence producing drugs and anti-Parkinson medication. The midbrain DA neurones are mainly located in the substantia nigra pars compacta (SNPC) and the ventral tegmental area (VTA). LÄS MER

  5. 5. Experimental studies of N-methyl-D-aspartate receptor NR3 subunits

    Författare :Anna Öhman; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : This thesis focuses on the NR3A and NR3B subunits of the excitatory glutamate receptor N-methyl-D-aspartate (NMDA). Functional NMDA receptors are believed to be tetrameric complexes with combinations of NR1 and NR2 subunits, and possible addition of NR3 subunits. LÄS MER
