1. Complement aberrations and autoantibodies to complement proteins in relation to disease mechanisms
Sammanfattning : The complement system, a part of the innate immune system with several links to the adaptive immune system, plays an important role in the pathogenesis of many diseases. The purpose of this thesis was to document and clarify some of these mechanisms. The thesis is based on four papers (I-IV). LÄS MER
2. C-reactive protein (CRP) and anti-CRP autoantibodies in systemic lupus erythematosus : a study on the occurrence and clinical implications of anti-CRP antibodies and CRP-mediated complement activation
Sammanfattning : Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by production of a wide range of autoantibodies, multiple organ involvement and by local formation or tissue deposition of immune complexes (ICs) in the inflamed organs. In contrast to most systemic inflammatory conditions, and despite raised levels of pro-inflammatory cytokines, SLE flares are rarely reflected by elevated C-reactive protein (CRP), an important acute-phase reactant in man with homologs in vertebrates and several invertebrates. 3. Recognition molecules of the complement system: C1q and mannan-binding lectin in autoimmunity and immune defence
Sammanfattning : The C1 subcomponent C1q and the mannan-binding lectin (MBL) are recognition proteins of the complement system, and have similar structure. C1q binds to IgG and IgM and activates the classical pathway. MBL binds to carbohydrate structures and activates the lectin pathway. LÄS MER
4. Studies on hereditary C2 deficiency: Frequent occurrence of severe infections, atherosclerosis and rheumatological manifestations
Sammanfattning : The complement system is a part of the innate immunity and is essential in the defence against microorganisms. Hereditary C2 deficiency (C2D) is one of the most common complement deficiency states with an estimated prevalence of 1:20,000 in persons of Western descent. LÄS MER
5. Reactivity to collagen type II and C1q in rheumatic diseases
Sammanfattning : In rheumatoid arthritis (RA),other arthritides and in systemic lupus erythematosus (SLE) there is immunological reactivity to collagen type II (CII) and to complement component C1q. Parts of the CII and C1q molecules are structurally and immunologically related. LÄS MER
