1. Hydrophobic interactions of serpins
Sammanfattning : The proteins of the serpin family are primarily but not exclusively proteinase inhibitors, which share a common, similar structure. It is known that this structure undergoes major conformational changes upon cleavage in the serpin reactive site loop, and that these changes include rearrangements within the hydrophobic core of the molecule. 2. Using peptide-phage display to capture conditional motif-based interactions
Sammanfattning : This thesis explores the world of conditional protein-protein interactions using combinatorial peptide-phage display and proteomic peptide-phage display (ProP-PD). Large parts of proteins in the human proteome do not fold in to well-defined structures instead they are intrinsically disordered. LÄS MER
3. Conformational Studies of Biologically Active Peptides
Sammanfattning : In this work conformational studies have been performed on peptides from four different systems in an attempt to gain knowledge about their biological function. Circular dichroism (CD) spectroscopy and nuclear magnetic resonance (NMR) spectroscopy in combination with distance geometry and restrained molecular dynamics simulations were the methods used. 4. NMR studies of the amyloid beta-peptide
Sammanfattning : The Amyloid beta peptide (Ab) is related to Alzheimer’s disease and is suggested to be the molecular pathogenic species of the disease, probably through the neurotoxic effect of Ab oligomers. Here the results from biophysical studies of Ab and fragments thereof, are presented. LÄS MER
5. Substrate specificity of protein kinases studied with synthetic peptides
Sammanfattning : This thesis concentrates on the substrate specificities of protein kinases A and C (PKA, PKC). Synthetic peptides are used for that purpose, both in soluble and cellulose membrane-bound form. The, stereospecificity of both protein kinases was studied with D-amino acid containing synthetic peptides. LÄS MER
