  1. 1. Bombesin Antagonists for Targeting Gastrin-Releasing Peptide Receptor-Positive Tumors : Design, Synthesis, Preclinical Evaluation and Optimization of Imaging Agents

    Författare :Zohreh Varasteh; Anna Orlova; Roger Schibli; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Bombesin; Gastrin-releasing peptide receptor GRPR ; Antagonist; Radionuclide molecular imaging;

    Sammanfattning : This thesis is focused on the development, preclinical evaluation, and optimization of radiotracers for the detection of gastrin-releasing peptide receptor (GRPR)-expressing tumors. The work is divided into three distinct parts: (1) the development of bombesin (BN) antagonist (RM26)-based imaging radiotracers for the detection of GRPR-expressing tumors using different positron emission tomography (PET) and single photon emission computed tomography (SPECT) radionuclides (68Ga, 18F and 111In), (2) the establishment of a method to monitor the ligand-G protein-coupled receptor (GPCR) interaction in real time without requiring purification and stabilization of the receptors, and (3) the evaluation of radiopeptide structure-related factors (length of mini-PEG linker and composition of chelator for metal labeling) affecting the in vitro and in vivo characteristics of RM26-based tracers. 2. 2. Prostate cancer theranostics using GRPR antagonist RM26

    Författare :Bogdan Mitran; Anna Orlova; Vladimir Tolmachev; Ulrika Rosenström; Tony Lahoutte; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Gastrin-releasing peptide receptor GRPR ; Bombesin; Prostate-specific membrane antigen PSMA ; Antagonist; Radionuclide molecular imaging; Theranostics; Peptide receptor radionuclide therapy PRRT .;

    Sammanfattning : The malignant transformation of cells is often associated with an alteration of their molecular phenotype, resulting in overexpression of several cell surface proteins. Gastrin-releasing peptide receptor (GRPR) and prostate-specific membrane antigen (PSMA) are examples of such pro-teins that are expressed at a high density in prostate cancer. LÄS MER

  3. 3. Neuropeptides and neurotrophins in arthritis : studies on the human and mouse knee joint

    Författare :Ola Grimsholm; Sture Forsgren; Solbritt Rantapää-Dahlqvist; Elvar Theodorsson; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; neuropeptides; neurotrophins; bombesin gastrin-releasing peptide; substance P; brain-derived neurotrophic factor; synovial tissue; knee joint; rheumatoid arthritis; Anatomy; Anatomi;

    Sammanfattning : Neuropeptides, such as substance P (SP) and bombesin/gastrin-releasing peptide (BN/GRP), and neurotrophins are involved in neuro-immunomodulatory processes and have marked trophic, growth-promoting and inflammation-modulating properties. The impact of these modulators in rheumatoid arthritis (RA) is, however, unclear. LÄS MER

  4. 4. Origin, release and functional significance of the novel gastric hormone ghrelin

    Författare :Charlotta Dornonville de la Cour; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; toxicology; Farmakologi; pharmacognosy; pharmacy; Pharmacological sciences; bone metabolism; energy homeostasis; glucagon; insulin; secretion; pancreastatin; somatostatin; histamine; gastrin; gastric acid secretion; ghrelin; gastric motility; A-like cells; farmakognosi; farmaci; toxikologi;

    Sammanfattning : The novel gastric hormone ghrelin was discovered by virtue of its ability to release growth hormone (GH) via interaction with the so-called GH-secretagogue receptor (GHS-R). Earlier only synthetic agents were known to bind with the GHS-R, hence, ghrelin is the first identified endogenous ligand. LÄS MER

  5. 5. Regulation and measurement of brown adipose tissue blood flow

    Författare :Gustavo Abreu-Vieira; Jan Nedergaard; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; fysiologi; Physiology;

    Sammanfattning : Brown adipose tissue (BAT) is an organ specialized in macromolecule combustion in order to produce heat. Because of its high capacity to dissipate energy, it is currently among the best hopes for future treatments of obesity and diabetes. LÄS MER
