1. Pharmacokinetic-Pharmacodynamic Modelling of Anticancer Drugs : Haematological Toxicity and Tumour Response in Hollow Fibres
Sammanfattning : Established quantitative relationships between dose, plasma concentrations and response [pharmacokinetic-pharmacodynamic (PKPD) models] have a high potential in improving therapeutic indices of anticancer drug therapy and in increasing drug development efficiency. PKPD modelling is a helpful tool for characterising and understanding schedule dependence. LÄS MER
2. Integrating Efficacy and Toxicity in Preclinical Anticancer Drug Development : Methods and Applications
Sammanfattning : Preclinical testing is an important part of cancer drug development. The aim of this thesis was to establish and evaluate preclinical in vitro methods useful in the development of new anticancer drugs. In paper I, the development of non-clonogenic assays (FMCA-GM) using CD34+ stem cells for assessment of haematological toxicity was described. LÄS MER
3. Nucleic Acids as Drugs and Drug Targets With Focus on Anticancer Active Platinum Complexes and Small Interfering RNAs
Sammanfattning : To ensure that patients receive a safe and effective treatment, it is important to understand how drugs interact with and affect molecules within a cell. Here, evaluation of platinum based drugs and their ability to interfere with nucleic acid functions is presented. LÄS MER
4. Analysis of drugs and proteins in biological matrices, using different types of sample preparations and mass spectrometry
Sammanfattning : This thesis describes bioanalysis of small and large molecules in biological matrices and includes screening of illegal drugs in urine with high resolution mass spectrometer, bioanalysis of MTH1 inhibitors in mice plasma and quantification of proteins in plasma and cell lysates.Screening of illegal drugs was based on high resolving power mass spectrometer (QTOF) and the results were compared to immunoassays. 5. Anticancer Activity of Melflufen : Preclinical Studies of a Novel Peptidase-Potentiated Alkylator
Sammanfattning : Melflufen (melphalan flufenamide, chemical name L-melphalanyl-p-L-fluorophenylalanine ethyl ester hydrochloride, previously called J1) is a derivative of the classical alkylating agent melphalan. Melflufen is potentiated by hydrolytic cleavage by aminopeptidase N (APN), leading to high intracellular concentrations of alkylating moieties and subsequent cell death. LÄS MER
