  1. 11. Development of HIV-1 Protease Inhibitors and Palladium-Catalyzed Synthesis of Aryl Ketones and N-Allylbenzamides

    Författare :Linda Axelsson; Mats Larhed; Anders Karlén; Luke Odell; Morten Grötli; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; palladium; aminocarbonylation; aryl ketones; decarboxylation; HIV; protease inhibitor; tertiary alcohol; macrocycles; Medicinal Chemistry; Läkemedelskemi;

    Sammanfattning : The use of palladium-catalyzed reactions to introduce new carbon-carbon bonds is a fundamental synthetic strategy that has been widely embraced due to its high chemo- and regioselectivity and functional group tolerance. In this context, Pd(0)-catalyzed aminocarbonylations using Mo(CO)6 instead of toxic and gaseous CO and with allylamine as the nucleophile were investigated. LÄS MER

  2. 12. Towards the Development of Photoswitchable β-Hairpin Mimetics

    Författare :Máté Erdélyi; Adolf Gogoll; Anders Karlén; John A. Robinson; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Organic chemistry; β-hairpin; peptidomimetics; Sonogashira; NMR; photochemistry; microwave; peptide; palladium; Organisk kemi; Organic chemistry; Organisk kemi;

    Sammanfattning : Peptide secondary structure mimetics are important tools in medicinal chemistry, as they provide analogues of endogeneous peptides with new physicochemical and pharmacological properties. The β-hairpin motif has been shown to be involved in numerous physiological processes, among others in regulation of eucariotic gene transcription. LÄS MER

  3. 13. Design and Synthesis of Enzyme Inhibitors Against Infectious Diseases : Targeting Hepatitis C Virus NS3 Protease and Mycobacterium tuberculosis Ribonucleotide Reductase

    Författare :Johan Gising; Mats Larhed; Anders Karlén; Victor Snieckus; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Medicinal Chemistry; Läkemedelskemi;

    Sammanfattning : Infectious diseases, including hepatitis C and tuberculosis, claim the lives of over 15 million people each year. Hepatitis C is caused by the hepatitis C virus (HCV) which infects the liver and can ultimately result in liver transplantation. HCV is very adaptive as a result of its high mutation rate. LÄS MER

  4. 14. Synthesis of Molecular Probes for Exploring the Human Consciousness, 5-HT7 Ligands and Salvinorins

    Författare :Pär Holmberg; Anders Karlén; Adolf Gogoll; Anette Johansson; Christer Sahlberg; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutical chemistry; serotonin system; opioid system; selective 5-HT7 agonist; 3-aminchromans; salvinorin A; selective kappa opioid receptor; mental disorders; consciousness; regioselective oxidation; enzymatic kinetic resolution; Farmaceutisk kemi; Pharmaceutical chemistry; Farmaceutisk kemi;

    Sammanfattning : In this study, we have addressed the serotonergic and the opioid system within the CNS. Both systems are of outmost importance in the etiology of disease states, especially mental disorders. LÄS MER

  5. 15. Structure-Based Virtual Screening : New Methods and Applications in Infectious Diseases

    Författare :Micael Jacobsson; Anders Karlén; Mats Kihlén; Antti Poso; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; : drug discovery; docking; scoring; virtual screening; malaria; tuberculosis; Pharmaceutical chemistry; Läkemedelskemi;

    Sammanfattning : A drug discovery project typically starts with a pharmacological hypothesis: that the modulation of a specific molecular biological mechanism would be beneficial in the treatment of the targeted disease. In a small-molecule project, the next step is to identify hits, i.e. molecules that can effect this modulation. LÄS MER
