1. Proislet Amyloid Polypeptide (proIAPP) : Impaired Processing is an Important Factor in Early Amyloidogenesis in Type 2 Diabetes
Sammanfattning : Amyloid is defined as extracellular protein aggregates with a characteristic fibrillar ultra-structure, Congo red affinity and a unique x-ray diffraction pattern. At present, 25 different human amyloid fibril proteins have been identified, and amyloid aggregation is associated with pathological manifestations such as Alzheimer’s disease, spongiform encephalopathy and type 2 diabetes. 2. Conjugated Polymers, Amyloid Detection and Assembly of Biomolecular Nanowires
Sammanfattning : The research field of conjugated polymers has grown due to the optical and electronic properties of the material, useful in applications such as solar cells and printed electronics, but also in biosensors and for interactions with biomolecules. In this thesis conjugated polymers have been used in two related topics; to detect conformational changes in proteins and to assemble the polymers with biomolecules into nanowires. LÄS MER
3. Amyloid aggregates: detection and interaction
Sammanfattning : The research on protein aggregation and amyloid formation is motivated by the fact that amyloid formation in tissue is harmful and associated with several debilitating diseases including Alzheimer’s disease (AD) and systemic amyloidosis such as transthyretin (ATTR) amyloidosis. Nevertheless, their beneficial roles in Nature have recently been identified, and artificial self-assembling of amyloid structure for various applications are emerging. LÄS MER
4. Bioanalysis using capillary electrophoresis and mass spectrometry : Applied on proteins, protein nanofibrils and polyvinyl alcohol microbubbles
Sammanfattning : The sequencing of the genome of various species, including the human species, have led to increased understanding about how a protein structure is generated, and how specific structures are related to the proteins’ functionality. In paper I and II of this thesis, the folding of proteins in vitro to form hierarchical nanostructures, which in vivo often have a pathological effect, have been studied. LÄS MER
5. Characterization of [18F]flutemetamol binding properties : A β-amyloid PET imaging ligand
Sammanfattning : The criteria for diagnosing Alzheimer’s disease (AD) have recently been revised to include the use of biomarkers for the in vivo presence of β-amyloid, one of the neuropathological hallmarks of AD. Examples of such biomarkers are positron emission tomography (PET) β-amyloid specific ligands, including [18F]flutemetamol. LÄS MER
