  1. 1. Alphavirus vectors as recombinant vaccines

    Författare :Peter Berglund; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Semliki Forest virus; Alphavirus; Synthetic vaccmes; Influenza A virus; HIV-I; SIV; DNA vaccines; AIDS vaccines; Influenza vaccine;

    Sammanfattning : This thesis describes further developments of an expression system based on the alphavirus Semliki Forest virus (SFV), and its potential use as recombinant vaccine. The RNA genome of SFV contains a 3'open reading frame (ORF) encoding the structural proteins, and a 5' ORF coding for the viral replicase. LÄS MER

  2. 2. The dynamic envelope of a fusion class II virus : molecular reorganizations during prefusion stages of Semliki forest virus

    Författare :Lars Haag; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Alphavirus; SFV; fusion; cryo-EM; formaldehyde; reorganization; cross-link; 3D reconstruction; acid pH; Pepscan; MAb; inactivation; dissociation; monoclonal antibody;

    Sammanfattning : The aim of this study was to explore a membrane fusion mechanism, prevailing in alphaviruses and known as virus class 11 fusion. The model virus for this mechanism is Semliki Forest virus (SFV) of the alphavirus family. LÄS MER

  3. 3. Genetic vaccination against acute viral disease

    Författare :Marina N Fleeton; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Semliki Forest virus; Alphavirus. Influenza A virus; DNA vaccines. Influenza vaccine. Respiratory Syncytial Viruses; Louping Ill. Flavivirus Infections;

    Sammanfattning : This thesis describes the development of recombinant vaccines based on the Semliki Forest virus (SFV) expression system. Immunisation of mice with recombinant virus particles, a layered DNA/RNA plasmid vector, and recombinant self-replicating RNA were carried out and the protective effect of these recombinant vaccines against viral challenge were examined.
