11. DNA checkpoint override and redox signaling in Schizosaccharomyces pombe
Sammanfattning : This thesis covers intracellular stress signaling through genotoxic stress, overriding of checkpoint control, as well as cellular redox status in hypoxic and oxidative stress Papers I and II: Caffeine has been shown to override cell cycle checkpoints in humans as well as in the fission yeast Schizosaccharomyces pombe. Understanding of the mechanism may aid in the development of compounds with similar overriding mechanisms for sensitization in cancer therapy. LÄS MER
12. Sequence- and structure guided engineering of proteins and enzymes for biotechnology and health applications
Sammanfattning : Proteins are highly diverse and sophisticated biomolecules that represent a cornerstone of biological structure and function and have been exploited in man-made applications for thousands of years. Those proteins that facilitate chemical reactions at physiologically relevant time-scales are referred to as enzymes. LÄS MER
13. Targeting the nucleotide metabolism of the mammalian pathogen Trypanosoma brucei
Sammanfattning : Trypanosoma brucei causes African sleeping sickness in humans and Nagana in cattle. There are no vaccines available against the disease and the current treatment is also not satisfactory because of inefficacy and numerous side effects of the used drugs.T. LÄS MER
14. Regulation of Ovarian Aromatase: Studies by Aromatase Assays in vitro and in vivo
Sammanfattning : An in vitro method was developed for measuring aromatase, based on binding of competitive aromatase inhibitor [11C]vorozole to the active site of the enzyme. [11C]Vorozole displayed high, specific binding in vitro to human placenta and human granulosa cells (GC), both fresh and frozen/thawed cells, provided correct procedures were used. LÄS MER
15. Reaction Mechanism and Allosteric Regulation of Class Ia Ribonucleotide Reductase from Escherichia coli
Sammanfattning : Ribonculeotide reductase (RNR) plays a key role in catalysing a reaction that provides all living organisms with building blocks for DNA. The RNR enzyme catalyses the conversion of ribonucleotides to their corresponding deoxyribonucleotides, by using free radical chemistry. LÄS MER
