    Sammanfattning : The human glucose transporter GLUT1 is abundant in red blood cells, the blood-brain barrier and epithelial cells, where it mediates the transport of the energy metabolite, glucose. In the present work some properties of GLUT1, including affinity binding of both substrates and inhibitors, transport rates as well as permeabilities of aromatic amino acids and drug-membrane interactions were analyzed by chromatographic methods.     Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Biochemistry; Affinity; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dissociation constant; Drug absorption; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Solute; Specific; Streptavidin; Wheat germ agglutinin; Biokemi; Biochemistry; Biokemi; Biokemi; Biochemistry;

    Sammanfattning : Specific and non-specific interactions of solutes with immobilized biomembranes were studied using chromatographic methods. Liposomes, proteoliposomes and red blood cell (RBC) membrane vesicles were immobilized by a freeze-thawing procedure, whereas whole RBCs were adsorbed in the gel beds using electrostatic interaction, binding to wheat germ agglutinin (WGA) or the streptavidin-biotin interaction. 4. 4. Selective extraction systems for detection and purification of membrane proteins

    Författare :Ulf Sivars; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; partitioning; Aqueous two-phase systems; separation; membrane proteins; detergent; micelle; polymer; Biochemistry; affinity; Metabolism; Biokemi;

    Sammanfattning : Increased understanding of membrane proteins is important for characterization of biochemical processes. Improved membrane protein isolation methods is a key issue for effective functional and structural determination of the large amount of unknown membrane proteins. LÄS MER

  5. 5. Affinity Two-Phase Partitioning of Liposomes and Membranes

    Författare :Irene Barinaga-Rementeria Ramírez; Biokemi och Strukturbiologi; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; miniaturised two-phase system; levitated drop; biotin; avidin; antibody; immunoaffinity; purification; rat lung; rat liver; caveolae; membrane; Affinity two-phase partitioning; liposome; Metabolism; Biochemistry; Biokemi; metabolism;

    Sammanfattning : Affinity two-phase partitioning based on an immunoaffinity sandwich approach for the rapid and selective purification of membranes is presented in this thesis. The method was developed by studying different parameters governing the affinity partitioning of model membranes. To this end, biotinylated liposomes were used. LÄS MER
