  1. 1. Cyclic Nucleotide Phosphodiesterase 3B: Regulation in rat adipocytes and 3T3-L1 cells

    Författare :Tova Landström; Institutionen för experimentell medicinsk vetenskap; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; 3T3-L1 TNF-alpha; adipocyte; insulin; p70S6 kinase; mitogen-activated protein kinase; protein kinase B; phosphatidylinositol-3 kinase; phosphodiesterase 3B; cAMP; lipolysis; phosphorylation; signal transduction; Biochemistry; Metabolism; Biokemi; metabolism;

    Sammanfattning : Insulin stimulation of rat adipocytes results in phosphorylation and activation of the cyclic nucleotide phosphodiesterase 3B (PDE3B), a key enzyme in the antilipolytic signalling pathway of this hormone. In this thesis, the site phosphorylated in PDE3B upon stimulation of rat adipocytes with insulin and/or the beta-adrenergic agonist, isoproterenol is identified. LÄS MER

  2. 2. Regulation of glycerol efflux in adipocytes. Structural and functional studies of the glycerol channel aquaporin 7

    Författare :Sofia de Mare; Medicinsk strukturbiologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Glycerol : metabolism; Lipolysis; X-ray Crystallography; Perilipin 1; Aquaglyceroporin; Aquaporin 7; Adipocytes metabolism; Glycerol : metabolism; adipocyte metabolism; X-ray crystallography; Aquaglyceroporin; Aquaporin 7; Perilipin 1; Lipolysis;

    Sammanfattning : Glycerol levels in adipocytes depend on the lipolysis, the hydrolysis of triglycerides into glycerol and free fatty acids, and the efflux of glycerol across the plasma membrane through glycerol channels. The aims of this work were to investigate how glycerol levels are regulated on a molecular level by PLIN1, a major lipid droplet-associated protein and a key regulator of the lipolysis by scaffolding for lipolytic proteins on the lipid droplet. Sammanfattning : Originally studying the molecular mechanisms for the insulin-like effects of growth hormone (GH), lipogenesis and antilipolysis, in isolated rat adipocytes we found that the GH receptor (GHR) was tyrosine phosphorylated in response to GH in cells that were responsive to these effects. Then, as Janus kinase 2 (JAK2) was described as a GH-stimulated and GHR-associated tyrosine kinase, we found that JAK2 was tyrosine phosphorylated in response to GH in responsive cells. LÄS MER

    Sammanfattning : Adipocytes are crucial energy reservoirs to maintain metabolic homeostasis of glucose and lipids in the human body.Glucose transporters (GLUTs) and aquaporins (AQPs) play an important role in metabolic regulation of glucose andlipids in human adipocytes. LÄS MER
