  1. 1. Pharmacogenetic Studies of Paclitaxel in Ovarian Cancer : focus on interindividual differences in pharmacodynamics and pharmacokinetics

    Författare :Henrik Green; Curt Peterson; Peter Söderkvist; Per Rosenberg; Håkan Melhus; Linköpings universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; pharmacogenetics; paclitaxel; LC MS; Taxol; ovarian cancer; pharmacokinetics; CYP2C8; CYP3A4; ABCB1; äggstockscancer; paklitaxel; Clinical pharmacology; Klinisk farmakologi;

    Sammanfattning : Ovarian cancer is one of the most common female cancer diseases in the world today and in Sweden more than 800 new cases are diagnosed every year. The standard treatment consists of chemotherapy with paclitaxel in combination with carboplatin after initial cytoreductive surgery. LÄS MER

  2. 2. ATP-Binding Cassette Efflux Transporters and Passive Membrane Permeability in Drug Absorption and Disposition

    Författare :Pär Matsson; Per Artursson; Ulf Norinder; Peter Swaan; Uppsala universitet; []
    Nyckelord :Pharmaceutics; ATP-binding cassette; ABC transporter; P-gp; P-glycoprotein; ABCB1; BCRP; Breast cancer resistance protein; ABCG2; MRP2; Multidrug-resistance associated protein 2; ABCC2; Membrane permeability; Drug transport; Active transport; Passive diffusion; Multivariate data analysis; PLS; OPLS; QSAR; Galenisk farmaci;

    Sammanfattning : Transport into and across the cells of the human body is a prerequisite for the pharmacological action of drugs. Passive membrane permeability and active transport mechanisms are major determinants of the intestinal absorption of drugs, as well as of the distribution to target tissues and the subsequent metabolism and excretion from the body. 3. 3. ATP-Binding-Cassette Transporters in Biliary Efflux and Drug-Induced Liver Injury

    Författare :Jenny M. Pedersen; Per Artursson; Christel A. S. Bergström; Stefan Oswald; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; ABC transport protein; Pgp; P-glycoprotein; ABCB1; BCRP; breast cancer resistance protein; ABCG2; MRP2; multidrug resistance-associated protein 2; ABCC2; BSEP; bile salt export pump; ABCB11; sandwich cultured human hepatocytes; SCHH; drug-induced liver injury; DILI; multivariate data analysis; OPLS; Galenisk farmaci; Pharmaceutics;

    Sammanfattning : Membrane transport proteins are known to influence the absorption, distribution, metabolism, excretion and toxicity (ADMET) of drugs. At the onset of this thesis work, only a few structure-activity models, in general describing P-glycoprotein (Pgp/ABCB1) interactions, were developed using small datasets with little structural diversity. 4. 4. Molecular heterogeneity of prostate cancer bone metastasis

    Författare :Erik Bovinder Ylitalo; Pernilla Wikström; Anders Bergh; Maria Brattsand; Vessela Kristensen; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Prostate cancer; CRPC; castration-resistant; androgen receptor; splice variant; abiraterone; cabazitaxel; ABCB1; cholesterol; Mdr1; P-gp; bone metastasis; immune response; metabolism; osteoblast; osteoclast; BMP; methylation;

    Sammanfattning : Castration-resistant prostate cancer (CRPC) develops after androgen deprivation therapy of advanced PC, often with metastatic growth in bone. Patients with metastatic CRPC have very poor prognosis. Growth of CRPC, in most but not all patients, seems to involve androgen receptor (AR) activity, despite castrate levels of serum testosterone. 5. 5. Interindividual Variability of Drug Transport Proteins : Focus on Intestinal Pgp (ABCB1) and BCRP (ABCG2)

    Författare :Gunilla Englund; Per Artursson; Håkan Melhus; Andreas Kindmark; Joseph W Polli; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Pharmaceutics; membrane transport proteins; ATP-Binding Cassette Transporters; P-glycoprotein; Breast Cancer Resistance Protein; Interindividual variability; Galenisk farmaci; Pharmaceutics; Galenisk farmaci;

    Sammanfattning : The appearance of adverse drug reactions is a common reason for hospitalization in Western countries. Research on underlying biological mechanisms for interindividual variability in drug response aims to better identify patients with exceptional genetic traits, disease conditions or risk of drug-drug interactions and thereby help to prevent adverse drug reactions.
