  1. 1. The Modular Domain Structure of ASF/SF2: Significance for its Function as a Regulator of RNA Splicing

    Författare :Vita Dauksaite; Göran Akusjärvi; Javier Caceres; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Molecular biology; pre-mRNA splicing; alternative splicing; splicing regulation; SR proteins; ASF SF2; modular domains; adenovirus; MLTU L1; E1A; MS2; Molekylärbiologi; Molecular biology; Molekylärbiologi;

    Sammanfattning : ASF/SF2 is an essential splicing factor, required for constitutive splicing, and functioning as a regulator of alternative splicing. ASF/SF2 is modular in structure and contains two amino-terminal RNA binding domains (RBD1 and RBD2), and a carboxy-terminal RS domain. LÄS MER

  2. 2. Functional Characterization of the Cellular Protein p32 : A Protein Regulating Adenovirus Transcription and Splicing Through Targeting of Phosphorylation

    Författare :Christina Öhrmalm; Göran Akusjärvi; Neus Visa; Uppsala universitet; []
    Nyckelord :Cell and molecular biology; Adenovirus; p32; ASF SF2; E4-ORF4; Splicing; Transcription; CTD; RNA Pol II; Cell- och molekylärbiologi;

    Sammanfattning : Cellular processes involved in the conversion of the genetic information from DNA into a protein are often regulated by reversible phosphorylation reactions. By modulating the phosphorylated status of key proteins their activity can either be enhanced or repressed. LÄS MER

  3. 3. Adenovirus vector systems permitting regulated protein expression and their use for in vivo splicing studies

    Författare :Magnus Molin; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Adenovirus; vector; inducible; gene expression; ASF SF2; splicing; Biokemi; Biochemistry; Biokemi; Medical Virology; medicinsk virologi;

    Sammanfattning : We have constructed two adenovirus-based gene expression vector systems permitting regulated protein expression. They are based on the tetracycline-regulated Tet-ON- and the progesterone antagonist RU 486-regulated gene expression systems, which were rescued into E1-deficient adenovirus vectors. LÄS MER

  4. 4. Viruses as a Model System for Studies of Eukaryotic mRNA Processing

    Författare :Anette Lindberg; Jan-Peter Kreivi; Göran Akusjärvi; Lars Wieslander; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Molecular biology; ICP27; herpes simplex virus; mRNA; splicing; adenovirus; ASF SF2; Molekylärbiologi; Molecular biology; Molekylärbiologi; Medical Virology; medicinsk virologi;

    Sammanfattning : Viruses depend on their hosts for the production and spread of new virus particles. For efficient virus replication, the viral genes have adapted the strategy of being recognized and processed by the cellular biosynthetic machineries. Viruses therefore provide an important tool to study the cellular machinery regulating gene expression. LÄS MER

  5. 5. Regulation of adenovirus alternative pre-mRNA splicing : Functional characterization of exonic and intronic splicing enhancer elements

    Författare :Bai-Gong Yue; Uppsala universitet; []

    Sammanfattning : Pre-mRNA splicing and alternative pre-mRNA splicing are key regulatory steps controlling geneexpression in higher eukaryotes. The work in this thesis was focused on a characterization of thesignificance of exonic and intronic splicing enhancer elements for pre-mRNA splicing. LÄS MER
