1. Selective Retention of β-Carbolines and 7,12-Dimethylbenz[a]anthracene in the Brain : Role of Neuromelanin and Cytochrome P450 for Toxicity
Sammanfattning : The ß-carbolines norharman and harman structurally resemble the synthetic compound 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) that is known for its ability to damage neuromelanin-containing dopaminergic neurons of the substantia nigra and thereby induce parkinsonism. MPTP is, however, not normally present in the environment whereas the ß-carbolines are present in cooked food and tobacco smoke. LÄS MER
2. Xenobiotic-metabolizing enzymes and enzymes involved in the metabolism of reactive oxygen species in the porcine ovary : subcellular distributions and levels of expression during follicular maturation and in the non-pregnant and pregnant corpus luteum
Sammanfattning : The polycyclic aromatic hydrocarbon (PAH) 7,12-dimethylbenz(a)anthracene (DMBA) is metabolized by both porcine ovarian microsomes and mitochondria, with the highest activity being seen in the corpus luteum. Microsomal DMBA monooxygenase activity is inhibited by inhibitors of the CYP450 system and by 17-bestradiol, thus indicating involvement of CYP1B1. LÄS MER
3. Bioactivation and binding of polycyclic aromatic hydrocarbons in blood vessel endothelia
Sammanfattning : Polycyclic aromatic hydrocarbons (PAHs) are widespread environmental pollutants formed by incomplete combustion of organic materials such as fossil fuels and wood. Experimental and epidemiological studies indicate that PAHs may be involved in the development of cardiovascular disease. LÄS MER
4. Adrenal Bioactivation and Toxicity of 3-MeSO2-DDE, o,p´-DDD and DMBA Investigated in Tissue Slice Culture
Sammanfattning : I developed a precision-cut adrenal slice culture procedure to investigate cytochrome P450 (CYP) catalysed irreversible binding and adrenocorticolytic effects in human, rodent, and fish adrenal tissue, ex vivo. Autoradiography and radioluminography of exposed tissue slices showed that the potent adrenal toxicant 3-methylsulphonyl-2,2´-bis(4-chlorophenyl)-1,1´-dichloroethene (MeSO2-DDE) causes a selective metabolite binding in zona fasciculata (ZF), which is diminished by the CYP11B1 inhibitor metyrapone. 5. Metabolism-dependent activation of food and environmental mutagens in endothelial cells
Sammanfattning : The endothelial cells of blood vessels have been proposed as a target for toxic effects ofxenobiotics in the cardiovascular system. In the present studies, induction of cytochromeP450 1A (CYP1A) enzymes and metabolic activation of food and environmentalmutagens were examined in endothelial cells of rodents, birds, and humans.
