  1. 1. Orientation and Conformation of Single Macromolecules on Unmodified and Functionalized Surfaces

    Författare :Magnus Bergkvist; David Cullen; Uppsala universitet; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biotechnology; Atomic Force Microscopy; protein orientation; protein adsorption; surface derivatization; Bioteknik; Bioengineering; Bioteknik; Surface Biotechnology; ytbioteknik;

    Sammanfattning : In this thesis methods for investigation of orientation and conformation of individual macromolecules on surfaces are presented as well as novel methods for functionalization of silicon chips with the possibility to get an ordered immobilization of antibodies. Two novel methods are presented which makes it possible to investigate the orientation of individual macromolecules on different kinds of surfaces with AFM. LÄS MER

  2. 2. Direct observation of biomolecule adsorption and spatial distribution of functional groups in chromatographic adsorbent particles

    Författare :Anders Ljunglöf; Abraham Lenhoff; Uppsala universitet; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biotechnology; Confocal microscopy; confocal Raman spectroscopy; protein adsorption; ligand distribution; visualization; imaging; Bioteknik; Bioengineering; Bioteknik; Surface Biotechnology; ytbioteknik;

    Sammanfattning : Confocal microscopy has been used as a tool for studying adsorption of biomolecules to individual chromatographic adsorbent particles. By coupling a fluorescent dye to protein molecules, their penetration into single adsorbent particles could be observed visually at different times during batch uptake. LÄS MER

  3. 3. Membrane-Assisted Isoform ImmunoAssay : Separation and determination of protein isoforms

    Författare :Maria Lönnberg; Carl G M Magnusson; Uppsala universitet; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biotechnology; Bioteknik; Bioengineering; Bioteknik; Surface Biotechnology; ytbioteknik;

    Sammanfattning : Proteins exist in a variety of isoforms with minor differences, mostly due to their glycosylation patterns, which can modulate their biological functions. It seems to be of clinical relevance to measure the isoform-distribution.Thesis describes a novel technology named Membrane-Assisted Isoform ImmunoAssay (MAIIA). LÄS MER

  4. 4. Endoglucanase and Mannanase from Blue Mussel, Mytilus edulis: Purification, Characterization, Gene and Three Dimensional Structure

    Författare :Bingze Xu; Folke Tjerneld; Uppsala universitet; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Biotechnology; Bioteknik; Bioengineering; Bioteknik; Surface Biotechnology; ytbioteknik;

    Sammanfattning : Two polysaccharide-degrading enzymes (endo-1,4-D-glucanase and β-mannanase) from blue mussel, Mytilus edulis, have been purified to homogeneity using a combination of several chromatographic steps. Each enzyme has been characterized with regard to its molecular weight, isoelectric point, pH and temperature stability, pH and temperature optimum and substrate specificity. LÄS MER

  5. 5. Development of Methods for Phase System Characterization in Liquid Chromatography

    Författare :Jörgen Samuelsson; Torgny Fornstedt; Karin Caldwell; Attila Felinger; Uppsala universitet; []
    Nyckelord :Analytical chemistry; Liquid chromatography; Adsorption isotherms; Adsorption energy distribution; Tracer pulse method; Elution by characteristic points; Peak deformation; Validation; Hybrid column material; Alkaline separation; Injection-profiles; Analytisk kemi;

    Sammanfattning : The aim of this thesis is first and foremost to improve the fundamental knowledge of nonlinear and preparative separation theory by focusing on some of the remaining “white spots” on the theoretical chromatographic map. Secondly, the acquired knowledge is used to develop, validate and execute new methods for phase characterization in liquid chromatography. LÄS MER
