  1. 1. Computational prediction of antisense oligonucleotides and siRNAs

    Författare :Alistair Chalk; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Antisense oligonucleotides; siRNAs; Artificial neural networks; Support vector machines; prediction; specificity.;

    Sammanfattning : Two popular gene knockdown methods, antisense oligonucleotides (AOs) and short interfering RNAs (siRNAs) are commonly used to selectively inhibit gene expression (gene knockdown). This is an extremely valuable tool for functional genomics, however the selection of effective molecules using either method is non trivial. LÄS MER

  2. 2. Molecular and diagnostic aspects of the protein p41 of HHV-6 and silencing of the CD46 receptor by RNA interference

    Författare :Yunhe Xu; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Human herpesvirus 6; p41; multiple sclerosis; RNA interference; small interfering RNAs; CD46;

    Sammanfattning : Multiple sclerosis (MS) is one of the most common diseases of the central nervous system (CNS), leading to sensory and motor disability in young adults. There is no efficient treatment of the disease. Its etiology is unknown. LÄS MER

  3. 3. Construction of Adenovirus Vectors for Studies of Protein Function and RNA Interference

    Författare :Saideh Berenjian; Göran Akusjärvi; Mikael Berg; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Microbiology; Adenovirus; viral vectors; VA RNAI II; RNAi; Tet-ON OFF; RNA splicing; Mikrobiologi; Microbiology; Mikrobiologi;

    Sammanfattning : During an adenovirus infection the accumulation of alternatively spliced mRNAs is subjected to a tight temporal regulation. The IIIa protein is a structural protein expressed exclusively late after infection. LÄS MER

  4. 4. Mechanistic characterization of post-transcriptional gene silencing

    Författare :Cecilia Dahlgren; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : With the ongoing task of identifying and characterizing genes in the human genome, there is a great demand for tools to study these genes. Today, reverse genetics is the most effective way to explore gene function. This involves suppression of the gene of interest by a set of manipulation techniques. LÄS MER

  5. 5. High resolution genomic tools for the discovery of protein function in mammalian cells

    Författare :Emily Carol Hodges; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Cell-based assays; overexpression; RNAi; BC2; C3F; Taxol;

    Sammanfattning : The work described in this thesis emphasizes the development of cell-based strategies with increased throughput design that enable rapid biological discovery through direct investigation of protein function. Each of the works represented in this thesis are unified by the common premise that genetic perturbations, through loss or gain-of-function, in cell-based models may serve as an entry point for assigning biological roles to uncharacterized genes and their gene products.
