    Sammanfattning : Abstract The present study evaluated genetic and metabolic factors influencing the risk for type 2 diabetes and cardiovascular disease (CVD) with special reference to increased concentrations of free fatty acids (FFA) in 1174 Swedish diabetic and nondiabetic subjects. Common polymorphisms in the fatty acid binding protein 2 (FABP2), the B2-adrenergic receptor (B2-AR), the B3 -adrenergic receptor (B3-AR) and the uncoupling protein 1 (UCP-1) genes were related to anthropometric and metabolic variables. Maternally Inherited Diabetes and Deafness (MIDD) and Maturity Onset Diabetes of the Young (MODY) are monogenic forms, whereas Latent Autoimmune Diabetes in Adults (LADA) is thought to be a polygenic subtype similar to T1D.     Sammanfattning : The aim of this thesis was to better understand the role of antigen presenting cells (APC) in the development of diabetes in the nonobese diabetic (NOD) mouse. We have shown that DC and Mphi that infiltrate the islets of Langerhans produce tumour necrosis factor-a (TNF-a), a cytokine known to contribute to the pathogenesis in the early stages of diabetes development.     Sammanfattning : The incidence of Diabetes Mellitus increases globally in epidemic proportions. Type 2 Diabetes is the most prevalent form of Diabetes, comprising 90% of the patients.
