1. On methods for probing structure and function of membrane proteins
Sammanfattning : The determination of structure–function relationships in membrane proteins plays a key role for the understanding of molecular mechanisms in biological systems, and may provide information on how these systems can be modulated by pharmaceuticals. The aim of the work described herein was to develop methods for studying structure and function of membrane proteins, ideally in combination. LÄS MER
2. Probing Proton Translocation in Influenza A/M2 Proteoliposomes - A systematic Approach to Membrane-Protein Reconstitutions
Sammanfattning : An improved method for reconstituting membrane proteins into artificial liposomes for quantitative functional analysis is presented. A number of key parameters for reconstitution by detergent removal are assessed in this thesis: The lipid-to-protein ratio, the detergent-to-lipid ratio and the lipid and cholesterol composition. 3. Chromatographic characterization of the human red cell glucose transporter Glut1 in the cells, in membrane vesicles and in proteoliposomes
Sammanfattning : The human glucose transporter Glut1 is used by many cells, especially red blood cells andbrain cell, to take up glucose and other necessary metabolites. Gentle chromatographic methods can be applied for analyses of Glut1 activities in lipid environments similar to the native one. LÄS MER
4. Drug Partitioning into Natural and Artificial Membranes : Data Applicable in Predictions of Drug Absorption
Sammanfattning : When drug molecules are passively absorbed through the cell membrane in the small intestine, the first key step is partitioning of the drug into the membrane. Partition data can therefore be used to predict drug absorption. LÄS MER
5. Chromatographic Studies of Solute Interactions with Immobilized Red Blood Cells and Biomembranes
Sammanfattning : Specific and non-specific interactions of solutes with immobilized biomembranes were studied using chromatographic methods. Liposomes, proteoliposomes and red blood cell (RBC) membrane vesicles were immobilized by a freeze-thawing procedure, whereas whole RBCs were adsorbed in the gel beds using electrostatic interaction, binding to wheat germ agglutinin (WGA) or the streptavidin-biotin interaction.
