1. The Multifunctional Pipette. A Microfluidic Technology for the Biosciences
Sammanfattning : The theme of the work described in this thesis is the generation and application of liquid microenvironments in chemistry and bioscience using microfluidic devices. First, a computer controlled multi-stage dilution system to generate time-dependent chemical waves was developed, and its application was demonstrated on model biomembranes. LÄS MER
2. Membrane-Polymer Interactions in Lipid Vesicles
Sammanfattning : Membrane related biological processes are commonly investigated in artificial biomimetic experimental systems. One of the most versatile models is based upon giant unilamellar phospholipid vesicles (liposomes), which are artificially generated spherical lipid structures in an aqueous environment. LÄS MER
3. Microfluidics based techniques for electrophysiological studies of cells
Sammanfattning : This thesis work investigates the application of microfluidics to perform electrophysiological studies on cells, including investigations of the effect of cholesterol on the dynamic ion permeability of TRPV1 ion channels, and the application of a microfluidic device, the multifunctional pipette, in electrophysiological studies on brain slices. In the first part of this thesis, Chinese hamster ovary (CHO) cells overexpressing the TRPV1 ion channel were used in a dynamic ion permeability study, where the activation properties of the TRPV1 ion channel were investigated using the patch clamp technique after depletion of membrane cholesterol. 4. A gastight microfluidic system combined with optical tweezers and optical spectroscopy for electrophysiological investigations of single biological cells
Sammanfattning : Stroke affects around 20 million people around the world every year. Clinically, stroke is a result of brain damage due to the shortage of oxygen delivered to the nerve cells. To minimize suffering and costs related to the disease, extensive research is performed on different levels. LÄS MER
5. Lab on a chip for electrophysiological measurements with control of the oxygen content : optical manipulation and spectroscopic analysis of biological cells
Sammanfattning : Stroke affects nearly 20 million people around the world every year. Clinically, stroke is a result of brain damage due to the shortage of oxygen delivered to the nerve cells. To minimize suffering and costs related to the disease, extensive research is performed on different levels. LÄS MER
