1. Activation of murine cytotoxic cells with interelukin-2 and the bacterial superantigen staphylococcal entertoxin A
Sammanfattning : Natural killer (NK) cells and T lymphocytes are the major effector cells that can recognize and kill tumor cells. NK cells have a constitutive cytotoxic activity and kill a wide spectrum of tumor cells while T cells recognize specific tumor antigens and need to be activated through their TCR to differentiate into cytotoxic T lymphocyte (CTL) killers or T helper cells. 2. Influence of FK506 on certain aspects of lymphocyte activation and lymphocyte-endothelial cell interactions in vitro
Sammanfattning : In the present thesis, the influence of the immunosuppressive drug FK506 on mitogen-induced changes in the expression of subunits a and ß of the IL-2 receptor by isolated human peripheral blood mononuclear cells has been studied. Up-regulation and release of the a-chain were potently depressed by FK506, even in the presence of exogenous IL-2. LÄS MER
3. Regulatory mechanisms controlling T cell activity
Sammanfattning : This thesis is based on work studying the immunoregulatory mechanisms that govern the balance between activation and inactivation of T lymphocytes. Most of the work has been carried out in an experimental system using a T cell receptor (TCR-Vb3) transgenic mouse strain to obtain a homogenous T-cell population and utilizing the properties of bacterial superantigens to induce T cell responses and unresponsiveness in vivo. 4. Genetic modification of human natural killer cells and possible applications thereof
Sammanfattning : Natural killer (NK) cells are dependent on the presence of interleukin-2 (IL-2) for their survival and for their cytotoxicity against tumor cells. Currently, for in vitro expansion of NK cells from peripheral blood mononuclear cells (PBMCs), IL-2 is of great importance. LÄS MER
5. Islet xenotransplantation : An immunological study in the pig-to-mouse model
Sammanfattning : Successful clinical xenotransplantation, i.e., transplantation between species, would eliminate the shortage of donor organs. In order to study the acute cellular rejection reaction following discordant xenogeneic transplantation, an experimental pig-to- mouse islet xenotransplantation model was established. LÄS MER
