21. Arrayed identification of DNA signatures
Sammanfattning : In this thesis techniques are presented that aim to determine individual DNA signatures by controlled synthesis of nucleic acid multimers. Allele-specific extension reactions with an improved specificity were applied for several genomic purposes. Since DNA polymerases extend some mismatched 3’-end primers, an improved specificity is a concern. LÄS MER
22. Molecular cytogenetic investigations of chromosomal abnormalities in prostate and urinary bladder cancers
Sammanfattning : Tumorigenesis is suggested to be caused by base pair mutations and chromosomal abnormalities leading to inactivation of tumor suppressor genes an activation of oncogenes. However, still it is not clear for the vast majority of these changes how they contribute to the tumorigenesis of the prostate and bladder cancers. LÄS MER
23. Interphase cytogenetics and cytokine therapy in myelodysplastic syndromes and acute myeloid leukemia
Sammanfattning : Myelodysplastic syndromes (MDS) comprise a group of clonal hematopoietic bone marrow cell disorders, characterized by a variable degree of anemia, granulocytopenia and/or thrombocytopenia and with a substantial risk of transformation to acute myeloid leukemia (MDS-AML). In the present thesis a method based on fluorescence in situ hybridization (FISH) was developed which allows cytogenetic studies on morphological identified peripheral blood and bone marrow cells (MGG/FISH). 24. Analysis of chromosomal rearrangements and gene copy number changes in breast cancer cells
Sammanfattning : Genome changes in terms of numerical chromosomal aberrations and structural rearrangements, including deletions, amplifications and translocations, gradually accumulate in the genome during tumor development. These genomic changes are likely to play an important role in the process of tumor progression. LÄS MER
25. A microcell hybrid based 'elimination test' in the search for malignancy related genes
Sammanfattning : This thesis is focused on the elimination test developed by us (Imreh et al, 1994). The elimination test was designed as a functional test for malignancy related gene identification. Its task is to follow the cytogenetic and molecular alterations on single normal chromosomes transferred into tumor cells. LÄS MER
