  1. 11. The pathophysiology of respiratory chain dysfunction

    Författare :José Pablo Silva; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mitochondrial disease; mitochondrial DNA mtDNA ; mitochondrial transcription factor A Tfam ; Cre-loxP; conditional knockout; reactive oxygen species ROS ; superoxide; uncoupling protein UCP ; mitochondrial bioenergeties; energy expenditure;

    Sammanfattning : Mutations of mitochondrial DNA cause a variety of clinical syndromes. It is unclear whether impaired oxidative phosphorylation on its own is the main cause of pathology or whether other factors such as secondary metabolic alterations, enhanced formation of reactive oxygen species (ROS) and induction of apoptosis also may contribute to the clinical phenotype. 2. 12. Gene expression during hard tissue formation

    Författare :Catharina Ellerström; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :bone; dentin; mRNA; fibrillar collagens; osteonectin; fibronectin; development; rat;

    Sammanfattning : The purpose of this thesis was to study the formation of the extracellular matrix (ECM) of bone and dentin during development. Molecular techniques, such as in situ hybridization and RT-PCR, were used to investigate the occurrence of specific gene products in formative cells. LÄS MER

  3. 13. Structural characterisation of mitochondrial macromolecular complexes using cryo-EM : Mitoribosome biogenesis and respiratory chain supercomplex

    Författare :Sorbhi Rathore; Martin Ott; Daniel Wilson; Stockholms universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; Mitochondria; mitoribosome biogenesis; mitoribosome assembly factors; yeast respiratory supercomplexes; single particle electron cryo-microscopy; cryo-EM sample preparation; Biochemistry; biokemi;

    Sammanfattning : Mitochondria, popularly known as powerhouse of the cell, contain specialised mitoribosomes that synthesise essential membrane proteins. These essential proteins are required to form enzyme complexes, which carry out the process of oxidative phosphorylation (OXPHOS). LÄS MER

  4. 14. Cellular responses to respiratory chain dysfunction

    Författare :Anna Hansson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Mitochondria; TFAM; cardiomyopathy; aging; reactive oxygen species ROS ;

    Sammanfattning : Mitochondria are network-like organelles present in most mammalian cells. They contain the respiratory chain (RC) that produces the majority of the energy needed in the cell, in the form of ATP. RC deficiency can arise due to mutations in the mitochondrial DNA (mtDNA) or in nuclear genes encoding mitochondrial proteins. LÄS MER

  5. 15. Structure formation at solid/liquid interfaces : Understanding self-assembly and environmental challenges

    Författare :Shirin Nouhi; Adrian R. Rennie; Vassilios Kapaklis; Maja S. Hellsing; Ann Terry; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; Solid liquid interface; neutron scattering; colloidal particles; self-assembly; perfluoroalkyl substance; Moringa seed proteins; adsorption; lamellar disorder; thermal fluctuations.;

    Sammanfattning : The work described in the present dissertation has explored the structure of particles and molecules at solid/liquid interfaces, aiming to understand the physics of self-organizing systems and use this knowledge to address some environmental issues. Surface-sensitive neutron scattering techniques, such as reflectometry and grazing incidence small angle scattering, have been used as the primary tool to investigate structures in proximity to an interface.
