  1. 1. Pathogenic effects of alpha-synuclein: modifications and toxicity

    Författare :Natalie Landeck; Brain Repair and Imaging in Neural Systems (BRAINS); []
    Nyckelord :Parkinson s disease; adeno-associated viral vector; animal models; alpha-synuclein; substantia nigra; dopaminergic neurons; phosphorylation; aggregation; toxicity;

    Sammanfattning : Accumulation of intraneuronal inclusions, containing mainly a protein called alpha-synuclein (asyn), is thought to play a critical role in neurodegenerative disorders such as Parkinson's disease (PD). In these so called Lewy bodies, the majority of asyn is phosphorylated at serine 129 (pS129) suggesting that post-translational modifications may play a critical role in the process of aggregate formation. 2. 2. Gene therapy by enzyme replacement for Parkinson’s disease Optimization of continuous DOPA delivery and development of a candidate vector for clinical application

    Författare :Tomas Björklund; Brain Repair and Imaging in Neural Systems (BRAINS); []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; serotonin; Parkinson’s disease; L-DOPA; dyskinesia; GTP cyclohydrolase 1; tyrosine hydroxylase; dopamine; gene therapy; enzyme replacement; gene regulation; aromatic L-amino acid decarboxylase;

    Sammanfattning : The introduction of L-DOPA pharmacotherapy revolutionized the treatment of Parkinson’s disease close to fifty years ago. Before the discovery of dopamine as a neurotransmitter and its involvement in the disease, these patients had very little aid from medication and were bound to a life without movement. LÄS MER

  3. 3. Advancing gene therapy for epilepsy. Translational pre-clinical studies with neuropeptide Y and glial cell-line derived neurotrophic factor

    Författare :Esbjörn Melin; Epilepsicentrum; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Epilepsy; Gene Therapy; neuropeptide Y NPY ; Y2 receptor; GDNF; Encapsulated cell biodelivery; seizure; In-vivo;

    Sammanfattning : Epilepsy is a neurological disorder, affecting approximately 1% of the population. The high rate of drug-resistance in epilepsy in general (30%), and temporal lobe epilepsy (TLE) in particular, pose a serious clinical problem. Thus, novel treatments for drug-resistant epilepsy is highly warranted. LÄS MER

  4. 4. Progressive models of Parkinson's disease : behavioural and neurochemical analyses

    Författare :Alexandra Alvarsson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Parkinson’s disease (PD) is a common neurodegenerative disorder diagnosed based on the presence of motor symptoms, including resting tremor, postural impairment, bradykinesia and rigidity. The pathological hallmarks are degeneration of midbrain dopaminergic neurons and the accumulation of inclusions containing the misfolded protein α-synuclein (termed “Lewy bodies”) throughout the nervous system. 5. 5. Serotonin in emotional memory processes : neuropharmacological studies with emphasis on depression

    Författare :Therese M Eriksson; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :;

    Sammanfattning : Cognitive dysfunctions are common in neuropsychiatric disorders including Major Depressive Disorder, but are difficult to normalize by antidepressant therapies and contribute to reduced functional improvement. Cognitive impairments include alterations of emotional memory processes that are early targeted by antidepressant therapies, including Selective Serotonin Reuptake Inhibitors (SSRIs). LÄS MER
