Sammanfattning : Altered transport of the catecholamines and serotonin precursor amino acids tyrosine and tryptophan, might be one explanation for the dysfunctional neurotransmission implicated in the pathophysiology of bipolar disorder and Attention Deficit/Hyperactivity Disorder (ADHD). In previous studies, an altered amino acid transport has been found in schizophrenia and autism, when using the fibroblast cell model. LÄS MER
4. Targeting double-stranded DNA with locked nucleic acid-based (LNA) oligonucleotides (ON) and DNA-binding peptide conjugates
Sammanfattning : In this thesis we have studied two different types of molecules that bind to the major groove of the DNA: (i) oligonucleotides (ONs) containing locked nucleic acid (LNA) and in combination with intercalating agents (Twisted Intercalating Nucleic Acid, TINA, and Benzoquinoquinoxaline, BQQ) and (ii) peptides mimicking the leucine zipper transcription factor (GCN4 bZIP TF). Both ONs and peptides bind to the DNA double helix aiming to serve as transcription regulators of disease related genes. LÄS MER
