1. Characterization of artificial and biological lipid vesicles using TIRF and SPR
Sammanfattning : Synthetic lipid vesicles serve as important mimics of cells and the natural membranes that they are enclosed by. As such they are frequently used as simplified models of the highly complex cell membrane to aid in-depth physicochemical and biological characterization of this essential biological structure. 2. Photo-induced dark states influorescence spectroscopy – investigations & applications
Sammanfattning : This thesis focuses on investigations of transient dark states of fluorescentmolecules using spectroscopic techniques. The main purpose is to show andconvince the reader that transient dark states are not always a nuisance, butalso represent an additional source of information. LÄS MER
3. Fluorescence microscopy studies of interactions between monomeric α-Synuclein and lipid membranes
Sammanfattning : α-Synuclein (α-Syn), is an intrinsically disordered protein present in the substantia nigra part of the brain. α-Syn is commonly known to associate with lipid membranes containing anionic lipids. LÄS MER
4. Transient State Fluorescence Microscopy - method development and biological applications : Exploiting the dark states of fluorophores to measure oxygen concentrations, redox state, Förster resonance energy transfer and membrane viscosity
Sammanfattning : Due to their long lifetime, triplet, redox and other transient states of fluorophores are highly sensitive to the micro-environment. Imaging their spatial distribution in biological samples can therefore help answer interesting questions about the metabolism, molecular interactions and dynamics in living cells. LÄS MER
5. DNA-Controlled Lipid-Membrane Fusion
Sammanfattning : Membrane fusion is essential for nerve-cell communication, for protein transport between cell organelles and the cell-membrane and for enabling the merger between virus and host membranes during virus infection. We have demonstrated that short DNA oligonucleotides, membrane-attached via CH in an orientation that mimics the overall zipperlike architecture of fusion-inducing proteins, induce fusion of both suspended vesicles and vesicles site-specifically tethered to SLBs.
