1. Redox balancing in recombinant strains of Saccharomyces cerevisiae
Sammanfattning : In metabolically engineered Saccharomyces cerevisiae expressing Pichia stipitis XYL1 and XYL2 genes, encoding xylose reductase (XR) and xylitol dehydrogenase (XDH), respectively, xylitol is excreted as the major product during anaerobic xylose fermentation and only low yields of ethanol are produced. This has been interpreted as a result of the dual cofactor dependence of XR and the exclusive use of NAD+ by XDH. LÄS MER
2. Protein-RNA interplay in translation. Structural studies of RRF, SelB and L1
Sammanfattning : Translation is a central biological process, that is catalysed by a machinery consisting of RNA and protein. In this thesis three different protein components of the bacterial protein synthesis system have been structurally studied. LÄS MER
3. Protein production, characterization and structure determination in structural genomics
Sammanfattning : This thesis covers the process from expression of a heterologous gene in Escherichia coli to structure determination of a protein by nuclear magnetic resonance (NMR) spectroscopy. The first part concerns structural genomics-related parallel screening studies on the effect of fusion tags (in particular the His tag) on protein solubility and the use of fusion tags in fast, parallel purification protocols intended for initial biophysical characterization of human proteins produced in E. 4. The GRE3 encoded aldo-keto reductase and its influence on xylose fermentation in recombinant Saccharomyces cerevisiae strains
Sammanfattning : The aim of the work presented in this thesis was to improve ethanol formation from xylose in recombinant Saccharomyces cerevisiae strains. This was achieved by decreasing the xylitol by-product formation. Gre3p is the major xylose-reducing enzyme in S. cerevisiae. LÄS MER
5. Proton transfer in nitric oxide reducing heme-copper oxidases
Sammanfattning : Heme-copper oxidases (HCuOs) are best known as terminal oxidases in the aerobic respiratory chain, in which they catalyze the reduction of oxygen to water. By receiving protons and electrons from opposite sides of the membrane as well as pumping protons, HCuOs contribute to the electrochemical proton gradient over the membrane that can be used for ATP synthesis.
