  1. 1. Colorectal Cancer : Aspects of Heredity, Prognosis and Tumour Markers

    Författare :Lana Ghanipour; Helgi Birgisson; Lars Påhlman; Karin Jirström; Tom Öresland; Uppsala universitet; []
    Nyckelord :colorectal cancer; heredity; Tryptophanyl t-RNA synthetase; microsatellite instability; SP-PLA; prognosis; biomarkers;

    Sammanfattning : Colorectal cancer (CRC) is one of the most common cancer types and leading causes of cancer death worldwide. Since CRC is a heterogenic disease, there is a demand for increased knowledge of the underlying genetic and epigenetic mechanisms. The aim of this thesis was to investigate heredity and potential tumour markers in relation to prognosis. LÄS MER

    Sammanfattning : Protein biomarkers circulating in blood hold the promise of improved diagnosis, prognosis and follow-up of treatment of disease via minimally invasive procedures. For the discovery and validation of such biomarkers, methods are needed that can facilitate parallel, highly specific and in-depth analysis of the blood proteome. LÄS MER

  3. 3. Proximity Ligation Assays for Disease Biomarkers Analysis

    Författare :Rachel Yuan Nong; Ulf Landegren; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; proximity ligation assay; blood biomarkers; protein interactions; pathway analysis; single molecule; next-generation sequencing; Molekylär medicin; Molecular Medicine;

    Sammanfattning : One of the pressing needs in the field of disease biomarker discovery is new technologies that could allow high performance protein analysis in different types of clinical material, such as blood and solid tissues. This thesis includes four approaches that address important limitations of current technologies, thus enabling highly sensitive, specific and parallel protein measurements. LÄS MER

  4. 4. Proximity Ligation and Barcoding Assays : Tools for analysis of proteins and protein complexes

    Författare :Di Wu; Masood Kamali-Moghaddam; Ulf Landegren; Ola Söderberg; Jörg Hoheisel; Uppsala universitet; []

    Sammanfattning : Proteins are fundamental structural, enzymatic and regulatory components of cells. Analysis of proteins, such as by measuring their concentrations, characterizing their modifications, and detecting their interactions, provides insights in how biological systems work physiologically or pathologically at the molecular level. 5. 5. Development and Application of Proximity Assays for Proteome Analysis in Medicine

    Författare :Felipe Marques Souza de Oliveira; Masood Kamali-Moghaddam; Ulf Landegren; Mats Gullberg; Janne Lehtiö; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Solid-phase proximity ligation assay; post-translational modifications; glycosylation; phosphorylation; Enzyme-linked immunosorbent assay; immunoassay and rolling circle amplification; Proximity Extension Assay; inflammation protein biomarkers; autoantibodies; autoimmune disease;

    Sammanfattning : Along with proteins, a myriad of different molecular biomarkers, such as post-translational modifications and autoantibodies, could be used in an attempt to improve disease detection and progression. In this thesis, I build on several iterations of the proximity ligation assay to develop and apply new adaptable methods to facilitate detection of proteins, autoantibodies and post-translational modifications.
