  1. 1. Central nervous system targets for micturition control

    Författare :Rikard Pehrson; Avdelningen för klinisk kemi och farmakologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; central nervous system; Micturition; 5-HT; GABA; tramadol; NAD-299; CGP62349; apomorphine; cerebral infarction; toxikologi; farmaci; farmakognosi; Farmakologi; toxicology; pharmacy; pharmacognosy; Pharmacological sciences;

    Sammanfattning : Normal urine storage and bladder emptying are controlled by the central (CNS) as well as the peripheral nervous system. Dysfunction of these mechanisms can lead to urinary incontinence. Urinary tract dysfunctions have symptomatically been treated mainly with drugs acting peripherally. LÄS MER

  2. 2. Positron emission tomography in serotonergic drug development

    Författare :Bengt Andrée; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :PET; 5-HT1A receptors; 5-HT2A receptors; D2 dopamine receptors; antidepressive drugs; antipsychotic drugs; pindolol; NAD-299; S 16924; ORG 5222; M100907; WAY; DWAY;

    Sammanfattning : The serotonin neurotransmission system has a central role in drug treatment of depression and anxiety and is also discussed in relation to treatment of schizophrenia. The benefits of presently available drugs are however limited by partial of poor response and side effects. LÄS MER

  3. 3. Serotonergic receptor subtypes in learning and memory : focus on 5-HT1A, 5-HT1B and 5-HT2A receptors

    Författare :Maria Lüttgen; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Serotonin; 5-HT1A receptor; 5-HT1B receptor; 5-HT2A receptor; acetylcholine; NMDA receptor; water maze; passive avoidance; septum; hippocampus; teaming; memory; immunohistochemistry; mRNA;

    Sammanfattning : The aim of this thesis was to study how serotonin (5-HT) receptor subtypes can influence hippocampus/amygdala-dependent teaming and memory using the water maze (WM) and the passive avoidance (PA) tasks in rodents, and to provide a cellular correlate to the behavioral results using histochemical methods. Glutamatergic neurons in the trisynaptic circuit of the hippocampus are believed to be important for spatial teaming and memory as well as synaptic plasticity. LÄS MER

  4. 4. Development of radioligands for the serotonergic neurotransmission system for use in positron emission tomography

    Författare :Johan Sandell; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :PET; Radioligand; Brain; 5-HT1A receptor; serotonin transporter;

    Sammanfattning : The serotonergic system is implicated in several neuropsychiatric disorders, such as depression, anxiety and schizophrenia. Several hypotheses on the pathophysiology of these disorders are based on the pharmacology of serotonergic drugs. Selective serotonin reuptake inhibitors (SSRIs) are widely used for the treatment of depression and anxiety. LÄS MER

  5. 5. Modulation of acetylcholine release by serotonergic 5-HT1A and 5-HT1B receptors : a microdialysis study in the awake rat

    Författare :Xiao Jing Hu; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Microdialysis; Acetylcholine; Serotonin; 5-HT1A receptor; 5-HT1B receptor; Frontal cortex; Ventral hippocampus;

    Sammanfattning : The aim of the thesis was to investigate the effects of the 5-HT1A receptor antagonist robalzotan (NAD-299) and the 5-HT1B receptor antagonist NAS-181 ((R)-(+)-2-(3-morpholinomethyl-2Hchromen-8-yl) oxymethyl-morpholine methanesulfonate) on cholinergic, glutamatergic and GABAergic neurotransmission in the rat brain in vivo. The extracellular levels of acetylcholine (ACh), glutamate (Glu) and GABA were monitored by microdialysis in the frontal cortex (FC) and ventral hippocampus (VHipp) in separate groups of awake rats.
