1. Experimental studies in brain tumours : with special regard to multidrug resistance and the ErbB-family
Sammanfattning : Primary brain tumours, and especially the most common form malignant gliomas, usually display a pronounced resistance to other treatment modalities when surgery fails to cure. Growth factors, such as EGF and its receptor, frequently amplified and overexpressed in malignant gliomas, and factors associated with multidrug resistance have been suggested to at least partially explain the poor outcome. 2. Expression of multidrug resistance genes and proteins and effect of selenite in anthracycline-resistant human tumor cell lines
Sammanfattning : Treatment failure due to development of drug resistance is a serious complication in cancer treatment and the major cause of death in these patients. This thesis has focused on to gain knowledge about the mechanisms of multidrug resistance and to establish in vitro methods to characterize the resistant tumor cell phenotype. LÄS MER
3. Exposure to xenobiotic chemicals disrupts metabolism, rhythmicity and cell proliferation in Drosophila melanogaster
Sammanfattning : Most species are constantly exposed to xenobiotic chemicals through multiple routes. Among all categories of xenobiotics, phthalates and bisphenols are two of the most widely used plasticizers and can be found in polyvinyl chloride (PVC) materials, medical devices and even drinking water. 4. Transport proteins in mammary epithelial cells : studies in murine (HC11) and bovine (BME-UV) cells - effects of prochloraz
Sammanfattning : Transporters localized in membranes of secreting mammary epithelial cells are involved in delivery of essential nutrients into milk. However, drugs and other xenobiotics may be substrates of these transporters and thus be actively secreted into milk, which may pose a health threat to breastfed infants and dairy consumers. LÄS MER
5. Pharmacokinetic aspects of the blood-brain barrier transport and equilibration of opioids studied with microdialysis in rats and mice
Sammanfattning : The blood-brain barrier (BBB) transport and equilibration of opioids with different physico-chemical properties and similar structures - codeine, morphine, morphine3-glucuronide (M3G) and morphine-6-glucuronide (M6G) - were investigated in rats and mice using microdialysis. The influences of active efflux pumps, P-glycoprotein (Pgp) and multidrug resistance protein (MRP1), on the BBB transport of morphine and M3G were studied. LÄS MER
