  1. 1. Structure of eukaryotic DNA polymerase epsilon and lesion bypass capability

    Författare :Nasim Sabouri; Erik Johansson; Lars Thelander; Ola Hammarsten; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; DNA polymerase epsilon; DNA replication; Okazaki fragment; Translesion synthesis; DNA lesion; dNTP; Biochemistry; Biokemi;

    Sammanfattning : To transfer the information in the genome from mother cell to daughter cell, the DNA replication must be carried out only once and with very high fidelity prior to every cell division. In yeast there are several different DNA polymerases involved in DNA replication and/or DNA repair. LÄS MER

  2. 2. Role of yeast DNA polymerase epsilon during DNA replication

    Författare :Isabelle Isoz; Erik Johansson; Juhani Syväoja; Umeå universitet; []
    Nyckelord :LANTBRUKSVETENSKAPER; AGRICULTURAL SCIENCES; DNA polymerase epsilon; eukaryotic DNA replication; fidelity; leading strand; Dpb2; Biochemistry and clinical chemistry; Biokemi och klinisk kemi;

    Sammanfattning : Each cell division, the nuclear DNA must be replicated efficiently and with high accuracy to avoid mutations which can have an effect on cell function. There are three replicative DNA polymerases essential for the synthesis of DNA during replication in eukaryotic cells. LÄS MER

  3. 3. Functional and structural properties of eukaryotic DNA polymerase epsilon

    Författare :Olga Chilkova; Erik Johansson; Per Elias; Umeå universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; eukaryotic DNA replication; DNA polymerase epsilon; protein-DNA interaction; Biochemistry; Biokemi;

    Sammanfattning : In eukaryotes there are three DNA polymerases which are essential for the replication of chromosomal DNA: DNA polymerase alpha (Pol alpha), DNA polymerase delta (Pol delta) and DNA polymerase epsilon (Pol epsilon). In vitro studies of viral DNA replication showed that Pol alpha and Pol delta are sufficient for DNA replication on both leading and lagging DNA strands, thus leaving the function of Pol epsilon unknown. 4. 4. Structural and biochemical basis for the high fidelity and processivity of DNA polymerase ε

    Författare :Rais Ahmad Ganai; Erik Johansson; Per Elias; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; DNA polymerase ε; Crystal structure; P domain; Pre-steady state kinetics; β-hairpin loop; Primer switching; Strand displacement; biokemi; Biochemistry;

    Sammanfattning : DNA polymerase epsilon (Pol ε) is a multi-subunit B-family DNA polymerase that is involved in leading strand DNA replication in eukaryotes. DNA Pol ε in yeast consists of four subunits, Pol2, Dpb2, Dpb3, and Dpb4. Pol2 is the catalytic subunit and Dpb2, Dpb3, and Dpb4 are the accessory subunits. LÄS MER

  5. 5. Mechanisms controlling DNA damage survival and mutation rates in budding yeast

    Författare :Jörgen Wiberg; Andrei Chabes; Per Sunnerhagen; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; dNTPs; ribonucleotide reductase; translesion synthesis; TLS polymerases; medicinsk biokemi; Medical Biochemistry;

    Sammanfattning : All living organisms are made of cells, within which genetic information is stored on long strands of deoxyribonucleic acid (DNA). The DNA encodes thousands of different genes and provides the blueprint for all of the structures and activities occurring within the cell. LÄS MER
