  1. 1. DNA Interactions with Chiral Polyaza-aromatic Ruthenium(II) Complexes

    Författare :Per Lincoln; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; phenanthroline; binding geometry; binding isotherm; DNA; dppz; linear dichroism; intercalation; ruthenium; cooperative binding;

    Sammanfattning : The binding to DNA of a series of chiral ruthenium complexes is studied with optical spectroscopy techniques, including isotropic absorption, linear dichroism (LD) and emission anisotropy as well as steady-state and time-resolved luminescence. The thesis focuses on the ìlight-switchî complex [Ru(phen)2dppz]2+ which binds to DNA by intercalation and luminesces when bound to DNA but not in pure buffer solution (phen=1,10-phenanthroline; dppz=dipyrido[3,2-a:2',3'-c]phenazine). 2. 2. Stability of bacterial DNA in relation to microbial detection in teeth

    Författare :Malin Brundin; Ulf Sjögren; Anders Johansson; David Figdor; Inge Fristad; Umeå universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Cell-bound DNA; cell-death; dentine; DNA binding affinity; DNA decomposition; DNA preservation; extracellular DNA; hydroxyapatite; PCR; polymerase chain reaction; odontologi; Odontology;

    Sammanfattning : The fate of DNA from dead cells is an important issue when interpreting results from root canal infections analysed by the PCR technique. DNA from dead bacterial cells is known to be detectable long time after cell death and its stability is dependent on many different factors. LÄS MER

  3. 3. Quantum Chemical Studies of Chemotherapeutic Drug Cisplatin : Activation and Binding to DNA

    Författare :Johan Raber; David van der Spoel; Paolo Carloni; Uppsala universitet; []
    Nyckelord :Quantum chemistry; cisplatin; quantum chemistry; DNA; density functional theory; activation; JM118; substitution reaction; Platinum; guanine; adenine; cytostatic drug; aquation; anation; activation; Kvantkemi;

    Sammanfattning : The serendipitous discovery of the potent cytotoxic properties of cisplatin brought about a revolution in the treatment of certain types of cancer, but almost fifty years later, there still remain unknown areas in the chemistry of cisplatin. There are questions regarding which form of the drug reaches its DNA target, or why certain DNA sequences are more preferred than others for reaction with cisplatin. LÄS MER

  4. 4. Groove-binding Unsymmetrical Cyanine Dyes for DNA Applications

    Författare :Maja Eriksson; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; polarized spectroscopy; bacteriophage T5; cryo-transmission electron microscopy; dynamic light scattering; DNA; intercalation; cyanine dyes; groove-binding; fluorescence spectroscopy; gel electrophoresis;

    Sammanfattning : This work focuses on interactions between DNA and a group of newly developed unsymmetrical cyanine dyes. The aim of the work was to discover fluorescent dyes that can be used in biophysical studies of natural DNA where minimum perturbation of the DNA-helix is essential, where a slow dissociation rate of the dyes is needed and where the ionic strength is an important parameter. 5. 5. Competitive-Binding Based Optical DNA Mapping - From Bacterial Plasmids to the Human Genome

    Författare :Vilhelm Müller; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; genetics; single-molecule; nanofluidics; DNA; antibiotic resistance; optical DNA mapping; plasmids; competitive binding; fluorescence microscopy; bacteria;

    Sammanfattning : Significant advances within the field of DNA sequencing have allowed us to study DNA at a level of detail that was previously impossible. However, dynamic genomic regions with a high degree of structural variations, while being linked to disease in humans and increased resistance to antibiotics, are still challenging to characterize.
