1. Quantum Chemical Studies of Chemotherapeutic Drug Cisplatin : Activation and Binding to DNA
Sammanfattning : The serendipitous discovery of the potent cytotoxic properties of cisplatin brought about a revolution in the treatment of certain types of cancer, but almost fifty years later, there still remain unknown areas in the chemistry of cisplatin. There are questions regarding which form of the drug reaches its DNA target, or why certain DNA sequences are more preferred than others for reaction with cisplatin. LÄS MER
2. Biomarkers of internal exposure/dose : Methods to quantify adducts to protein and DNA by LC/MS studied with benzo[a]pyrene and isocyanates
Sammanfattning : This thesis focuses on methods for quantification by liquid chromatography/mass spectrometry (LC/MS) of specific biomarkers for internal dose of chemicals which induce toxicity through their electrophilic reactivity. In vivo such compounds are short-lived, and could feasibly be measured as their reaction products (adducts) with biomacromolecules. LÄS MER
3. Nucleic Acids as Drug Targets -Interactions of Platinum Complexes, Polyamines, and siRNA with DNA and RNA Models
Sammanfattning : Drugs targeting nucleic acids were studied in DNA and RNA model systems. The effects were investigated with respect to structural and thermodynamic influence, kinetics, and biological function. The drugs employed in this thesis were anticancer active platinum(II) complexes, polyamine derivatives, and siRNA. LÄS MER
4. DNA adductomics: Method development and integration in biological effect monitoring
Sammanfattning : Environmental omics can provide informative biomarkers for identifying pathologies or physiological responses to environmental change. Nucleic acid adductomics, one of the newest omics techniques, is particularly well suited for assessing exposure and effects of environmental contaminants because the technique can capture modifications at the (epi)genome level. 5. High-performance liquid chromatography for analysis of 32P-postlabeled DNA adducts
Sammanfattning : The formation of DNA adducts, i.e. the covalent binding of chemicals and chemical groups to DNA, is believed to be an important step in chemical carciogenesis. DNA adducts are usually formed at very low levels which requires very sensitive methods to analyze them. LÄS MER
