  1. 1. Quantitative Aspects of Nanodelivery Across the Blood-Brain Barrier : Exemplified with the Opioid Peptide DAMGO

    Författare :Annika Lindqvist; Margareta Hammarlund-Udenaes; Markus Fridén; William Elmquist; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; blood-brain barrier; liposomes; nanocarriers; brain delivery; pharmacokinetics; modeling and simulation; microdialysis; opioid peptide; DAMGO; LC-MS MS; Farmaceutisk vetenskap; Pharmaceutical Science;

    Sammanfattning : The use of nanocarriers is an intriguing approach in the development of efficacious treatment for brain disorders. The aim of the conducted research was to evaluate and quantify the impact of a liposomal nanocarrier formulation on the brain drug delivery. LÄS MER

  2. 2. Long-term Effects of Opioids in the Treatment of Chronic Pain : Investigation of Problems and Hazards on Clinical, Biochemical, Cellular and Genetic Levels

    Författare :Annica Rhodin; Torsten Gordh; Fred Nyberg; Mats Stridsberg; Mats Enlund; Harald Breivik; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; opioid; chonic pain; long-term opioid treatment; Anaesthetics; Anestesiologi; Anestesiologi; Anaesthesiology;

    Sammanfattning : After two decades of liberal prescribing of opioids, there has been an increasing recognition of problems connected to the prolonged use of opioids for chronic pain. The aim of my thesis was to explore some consequences of long-term opioid treatment for chronic pain such as problematic opioid use, endocrine disorders, tolerance and genetic variations in pain and opioid response. 3. 3. Characterization of Substance P (SP) Aminoterminal SP (1-7) Binding in Brain Regions and Spinal Cord of the Male Rat : Studies on the Interaction with Opioid Related Pathways

    Författare :Milad Botros; Fred Nyberg; Tsukasa Sakurada; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biological research on drug dependence; Biologisk beroendeforskning;

    Sammanfattning : Binding sites for substance P(1-7), SP(1-7) have been identified and characterized for the first time in crude membrane fraction from rat CNS using tritiated ([3H]) SP(1-7) as tracer. These putative receptors were investigated in relation to their affinity for tachykinins, opioid peptides and sigma receptor ligands. LÄS MER

  4. 4. In vitro and postmortem studies of the brain opioid system : association to opiate dependence

    Författare :Parisa Zarnegar; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :Opioid system; brain; gene expression; amygdala; hippocampus; opiate dependence;

    Sammanfattning : Opioids such as heroin are highly addictive illicit drugs. The μ opioid receptor (MOR) plays a central role in mediating the effects of opiates and related opioid. LÄS MER

  5. 5. Central pain after spinal cord injury : experimental studies with special emphasis on pharmacological treatment

    Författare :Wei Yu; Karolinska Institutet; Karolinska Institutet; []
    Nyckelord :allodynia; central pain; ischemia; spinal cord injury; intrathecal; opioid; opioid receptors; selective opioid agonists; selective opioid antagonists; tolerance; dependence; clonidine; bovine chromaffin cells; encapsulation; resiniferatoxin;

    Sammanfattning : Injuries and dysfunctions of the central nervous system can cause central pain which, according to clinical experience, is often refractory to conventional treatments. The present thesis consists of experimental studies on the mechanisms and treatments of central pain using a well-established animal model of central pain after spinal cord injury. LÄS MER
