  1. 1. Laboratory Soft X-Ray Cryo Microscopy: Source, System and Bio Applications

    Författare :Emelie Fogelqvist; Hans Hertz; David Attwood; KTH; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; X-ray microscopy; X-ray optics; Instrumentation; Laser plasma; Cryofixation; Tomography; Tomographic reconstruction; Autophagy; Immune synapse; Physics; Fysik; Physics;

    Sammanfattning : Soft x-ray microscopes routinely perform high-resolution 3D imaging of biological cells in their near-native environment with short exposure times at synchrotron radiation facilities. Some laboratory-sized microscopes are aiming to make this imaging technique more accessible to a wider scientific community. LÄS MER

  2. 2. Development and Evaluation of Preparation Methods for the Analysis of Cell Membranes Using Time-of-Flight Secondary Ion Mass Spectrometry

    Författare :Jakob Malm; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; salt; cryofixation; TOF-SIMS; supported phospholipid bilayer; freeze drying; vacuum; cells; POPC; glutaraldehyde;

    Sammanfattning : The aim of this work was to evaluate and develop methods for preparing dry samples for TOF-SIMS analysis of cell membranes. For correct interpretation of the analysis data, an understanding of the effects that the preparation method has on the morphology and chemical structure of the sample must first be obtained. 3. 3. Methods in electron microscopic quantitative immunocytochemistry using adenohypophysis as a model

    Författare :Barbara Kniola; Linköpings universitet; []
    Nyckelord :MEDICINE; MEDICIN;

    Sammanfattning : Quantitative ultrastructural immunocytochemistry (QUIC) is based on the assessment of immunogold labeling over the surface of ultrathin resin-embedded sections. The density of specific labeling, expressed as the number of gold particles/mm2 of a specific area can only be used as an approximative estimation of antigen concentration in the object. LÄS MER
