1. Towards unbiased 3D reconstruction : in single-particle cryo-electron microscopy
Sammanfattning : Cryo-electron microscopy of freestanding molecules (single-particles) plays a pivotal role in the difficult and pressing challenge of determining the structures of large macromolecular complexes. Molecular volumes are generated by aligning large sets of randomly oriented two-dimensional (2D) projection images in three dimensions (3D) before reconstruction is performed using tomographic techniques. 2. Single-particle cryo-electron microscopy of macromolecular assemblies
Sammanfattning : In this thesis, single-particle cryo-electron microscopy (cryo-EM) was used to study the structure of three macromolecular assemblies: the two hemocyanin isoforms from Rapana thomasiana, the Pyrococcus furiosus chaperonin, and the ribosome from Escherichia coli. Hemocyanins are large respiratory proteins in arthropods and molluscs. LÄS MER
3. Insights into structure and dynamics of the AAA+ motor of magnesium chelatase
Sammanfattning : The insertion of Mg2+ into protoporphyrin IX represents the first committed step in the chlorophyll and bacteriochlorophyll biosynthetic pathways. The reaction is catalyzed by the multisubunit enzyme Mg-chelatase, which consists of three subunits, known as I (molecular weight ~40 kDa), D (~70 kDa), and H (~140 kDa). LÄS MER
4. Analytic and data-driven methods for 3D electron microscopy
Sammanfattning : The central theme of this thesis is theoretical and algorithmic aspects of 3D electron microscopy (3D-EM). In particular, the thesis explores three parts of this theme. The first part concerns analysis of forward operators that, compared to those traditionally used, better account for the wave properties of the imaging electron. LÄS MER
