  1. 1. Antibody Evolution and Repertoire Development

    Författare :Johan Lantto; Institutionen för immunteknologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Immunology; serology; transplantation; Immunologi; serologi; virus neutralization; trinucleotide repeat; insertion; immunoglobulin variable genes; human cytomegalovirus; glycoprotein B; deletion; antibody; antibody repertoire;

    Sammanfattning : Antibodies are key players of the immune system in higher vertebrates, which provide a defense against potentially lethal threats from the environment. Besides their importance in the immune defense, antibodies have a great potential as reagents in biological chemistry and diagnostics, and as therapeutic agents against both infectious diseases and cancer. 2. 2. MuSK Antibody(+) Versus AChR Antibody(+) Myasthenia Gravis : Clinical, Neurophysiological and Morphological Aspects

    Författare :Anna Rostedt Punga; Erik Stålberg; Hannu Kalimo; John Newsom-Davis; Uppsala universitet; []
    Nyckelord :Neurosciences; myasthenia gravis; MuSK antibody; AChR antibody; myopathy; single-fiber EMG; mitochondria; Swedish MG questionnaire; quality of life; Neurovetenskap;

    Sammanfattning : Myasthenia gravis (MG) is an autoimmune neuromuscular disorder that causes fluctuating muscle weakness. MG may be divided into an ocular form and a generalized form based on the involved muscles. Treatment differs between these different MG forms. LÄS MER

  3. 3. Engineering Antibody Specificity Using Combinatorial Antibody Libraries

    Författare :Jenny Boström; Chalmers tekniska högskola; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; antibody Antigen Interactions; Dual Specific Antibodies; Herceptin; Human Epidermal Growth Factor Receptor 2; Thermodynamic Analysis; therapeutic Antibody Engineering; Synthetic Phagedisplayed Antibody Libraries; Vascular Endothelial Growth Factor; Bacterial Display; trastuzumab;

    Sammanfattning : The research presented in this thesis investigates antibody specificity, antibody-antigen and protein-protein interactions using combinatorial antibody libraries, phage-display and bacterial display technology. One goal was to develop a method to generate antibodies that bind two protein antigens with high affinity using a single-antigen binding site, and to investigate the molecular and thermodynamic requirements for antibody multispecificity. Sammanfattning : As many genome sequencing efforts today are completed, we are now provided with the genetic maps for several organisms, including man. With these maps at hand, the scientific focus is now shifting towards investigations of the functionality of proteins. LÄS MER

  5. 5. Generation and evolution of human antibody repertoires

    Författare :Helena Persson; Institutionen för immunteknologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; specificity; Immunology; serology; transplantation; Immunologi; serologi; Biotechnology; Bioteknik; phage display; IgE; hapten; grass pollen allergen; diversity; antibody repertoire; antibody library; antibody evolution; affinity maturation; allergy;

    Sammanfattning : This thesis, which is based on five original papers, covers different aspects of antibody specificity and diversity, with the overall goal to understand the makeup of antibody repertoires and their functionality as they occur in vivo or as they are developed in the laboratory. Besides their importance in the adaptive immune response, antibodies have emerged as an invaluable tool in applications ranging from basic research to disease diagnostics and therapy. LÄS MER
