  1. 1. Protein engineering to explore and improve affinity ligands

    Författare :Martin Linhult; KTH; []
    Nyckelord :binding; affinity; human serum albumin HSA ; albumin-binding domain ABD ; affinity chromatography; deamidation; protein A; stabilization; Z-domain; capacity; protein G; cleaning-in-place CIP ; protein engineering; C2 receptor;

    Sammanfattning : In order to produce predictable and robust systems forprotein purification and detection, well characterized, small,folded domains descending from bacterial receptors have beenused. These bacterial receptors, staphylococcal protein A (SPA)and streptococcal protein G (SPG), possess high affinity to IgGand / or HSA. LÄS MER

  2. 2. Protein microarrays for validation of affinity binders

    Författare :Mårten Sundberg; Peter Nilsson; Jochen Schwenk; Anders Isaksson; Afshin Ahmadian; KTH; []
    Nyckelord :TEKNIK OCH TEKNOLOGIER; ENGINEERING AND TECHNOLOGY; Microarray; protein; antibody; antigen; affinity; validation;

    Sammanfattning : Is specificity an important issue regarding affinity reagents? What about the validation of affinity reagents today, is it good enough? This depends on the application and the producer of the reagent. Validation should be the most important marketing argument that can be found. LÄS MER

  3. 3. Insights into weak affinity antibody-antigen interactionsStudies using affinity chromatography and optical biosensor

    Författare :Magnus Strandh; Institutionen för immunteknologi; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Bioteknik; Biotechnology; immunosensor; biosensor; weak affinity chromatography; cross-reactivity; antibody-antigen interaction; weak affinity; biomolecular recognition; Immunology; serology; transplantation; Immunologi; serologi;

    Sammanfattning : Molecular recognition and interaction is fundamental for the function of biological systems. The properties of the interacting biomolecules dictate the type of forces involved and the strength and dynamics of the interaction. Many interactions are very strong whereas others exhibit weak affinity. LÄS MER

  4. 4. Exploring molecular interactions between polypeptide conjugates and protein targets : Manipulating affinity by chemical modifications

    Författare :Aleksandra Balliu; Lars Baltzer; Oliver Seitz; Uppsala universitet; []
    Nyckelord :NATURVETENSKAP; NATURAL SCIENCES; NATURVETENSKAP; NATURAL SCIENCES; polypeptide conjugates; protein targets; molecular recognition; affinity enhancement; conjugation;

    Sammanfattning : In this thesis molecular interactions between polypeptide conjugates and protein targets were investigated. Polypeptides were derivatized with small organic molecules, peptides and oligonucleotides. New strategies were developed with the aim to increase affinities for proteins of biological interest. 5. 5. Affinity-, Partition- and Permeability Properties of the Human Red Blood Cell Membrane and Biomembrane Models, with Emphasis on the GLUT1 Glucose Transporter

    Författare :Christine Lagerquist Hägglund; Per Lundahl; Jørgen Vinten; Uppsala universitet; []
    Nyckelord :MEDICIN OCH HÄLSOVETENSKAP; MEDICAL AND HEALTH SCIENCES; Biochemistry; Affinity; Aromatic amino acids; Binding; Biomembrane; Biotin; Chromatography; Cytochalasin B; Dihydrocytochalasin B; Dissociation constant; Drug absorption; Ethanol; Equilibrium; Glucose; GLUT1; Immobilization; Immobilized biomembrane affinity chromatography; Immobilized liposome chromatography; Interaction; Liposome; Membrane protein; Membrane vesicle; Partitioning; Phospholipid bilayer; Proteoliposome; Quantitative; Red blood cell; Specific; Streptavidin; Tyrosine; Tryptophan; Biokemi; Biochemistry; Biokemi;

    Sammanfattning : The human glucose transporter GLUT1 is abundant in red blood cells, the blood-brain barrier and epithelial cells, where it mediates the transport of the energy metabolite, glucose. In the present work some properties of GLUT1, including affinity binding of both substrates and inhibitors, transport rates as well as permeabilities of aromatic amino acids and drug-membrane interactions were analyzed by chromatographic methods.
